小鼠皮肤光生物过程的光学显微观察与分析

【摘要】 光与生命的相互作用是人类发展过程中的一个永恒主题,光美容是光与生物组织相互作用的一个主要的应用,近年来光美肤术风靡整个世界,但是其作用过程及光与皮肤相互作用的机制至今还没有得到正确地认识,在临床上,时时受到并发症的困扰,其并发症内部的机理也不为人所知。这也是作者开展这个课题的意义,本论文结合动物实验与数值模拟,在亚微米数量级上研究非消融性光子嫩肤的作用及其美容的效果以揭示光与皮肤组织相互作用过程产生的效果。 论文的内容主要包括以下五部分: 简要介绍了光美容的历程与现状,提出目前存在的问题,揭示本论文的选题意义及拟解决的方案,概括其主要研究内容。 讨论了光子嫩肤的作用,重点说明了选择性光热解的原理,描述皮肤老化形成的原因及其病理上的表现,为实验提供了理论基础。 简要介绍本论文实验仪器的特点、应用及其在本论文实验中的具体应用,通过预实验来说明这些仪器在本论文中应用的可行性。 开展使用光子嫩肤仪及Er:YAG激光对活体鼠皮相互作用的实验研究。对实验结果进行总结,分析光子嫩肤仪和Er:YAG激光作用下皮肤,特别是真皮层的改变情况。通过激光共聚焦显微镜、微分相干衬显微镜对作用不同参数的鼠皮在形态学上进行显微观察,通过对实验组和对照组样品病理切片进行组织形态学上观察作为辅助比较,用双光子显微镜测量在光作用前后二次谐波及荧光光谱的变化情况。 作为扩展研究,利用皮肤的一些经验值,用蒙特卡罗模拟运算在与实验相同参数下作一部分近似简化来得到生物组织体内的光能量分布及温度的变化。

【Abstract】 The interaction of light and life is a permanent topic during the process of human being development, one of which main application is photo-rejuvenation and laser cosmetology. In recent years, photon-rejuvenation is fashionable in the world, but the theoretical study and the interaction of photo skin were still so poor, and then sufferers and physicians were undergong by complication in clinic and the mechanism was unknown, which are the significance of the subject we have. This thesis was devoted to study the mechanism of the non-ablation photorejuvenation and evaluating the effect after treatments in submicrometer using the means of combining the animal experimental and numerical analysis, so as to disclosure the process of the interacting in photo-skin.The paper was componented of the five parts as follows: In part one, the development course of photo-rejuvenation technology, including the history and actuality, was depicted in brief. The problem in present also was pointed out and the significance of this dissertation was exposed out. The main research contents of the paper were summarized. In part two, the mechanism of the photo-rejuvenation was discussed, the main part of which was the selective photothermolysis, then the reason of the formation of aging skin and that display in pathology was described. This provided the theoretical basis for experiment research. The characteristics and applications of the apparatus using in the experiment was depicted in brief. And then author have discussed the feasibility of the apparatus in this thesis’s experiment through the pre-experiment. The experiment research, which major in interaction of intense pulse light source and Er:YAG laser in rat-vivo skin were made. The result of compared group and experiment group was analyzed and summarized. In detail, the rat skin after interaction by IPL, especially the change of the dermas was analyzed. The Morphologic changes of rat skin were observed using laser scanning confocal microscopy after irradiated by IPL in different parameters. And then the tissue sectioning were made, and stained with H.E dyes as the assistant compare. Two-photon microscopy analysises the changesof second harmanic generation and fluorescence spectrum between pre- and after light irradiated.As a supplementary study, the numerical simulation of optical energy and temperature distribution in rat skin were carried out, using the optical and thermal parameters of the skin which obtained by experiential value, and then which results was compared with that of the animal experiments.
