【摘要】 目的:采用免疫组化链霉素抗生素蛋白过氧化物(S-P)法,观察趋化因子CXC家族中基质细胞衍生因子-1(SDF-1)的唯一受体CXC类趋化因子受体4(CXCR4 )在非小细胞肺癌(NSCLC)原发灶及淋巴结转移灶内肿瘤细胞中的表达情况,探讨NSCLC肿瘤细胞中CXCR4表达的临床意义以及CXCR4的表达在局部淋巴结转移中的作用。方法:用兔抗人CXCR4单克隆抗体对未做治疗的41例NSCLC肿瘤原发灶组织、其中25例伴有局部淋巴结转移的转移淋巴结组织和20例非肺部疾患的正常肺组织按S-P法进行免疫组化染色,显微镜下观察细胞CXCR4的表达情况。结果: 41例NSCLC原发灶中肿瘤细胞CXCR4阳性表达38例,阴性表达3例,阳性率为92.68%。其中CXCR4阳性表达(+)9例(21.95%),CXCR4阳性表达(++)14例(34.15%),CXCR4阳性表达(+++)15例(36.59%)。20例正常肺组织细胞CXCR4阳性表达(+)2例,阴性表达18例,阳性率10%。不同性别、年龄、TNM分期、肿瘤组织学类型的NSCLC患者其原发灶中肿瘤细胞CXCR4表达没有差异(P>0.05),同时CXCR4表达的强度与淋巴结转移情况也没有发现相关性(P>0.05)。25例局部转移淋巴结内肿瘤细胞CXCR4阳性表达24例,阳性率96%。其中CXCR4阳性表达(+)8例(32%),CXCR4阳性更多还原

【Abstract】 Objective: Using the method of Streptavidin-Peroxidase(S-P )immunohistochemical staining to observe the expression of the unique receptor, which is named as chemokine CXC motif receptor 4(CXCR4) to stromal cell-derived factor-1(SDF-1) in primary tumor cells of non small cell lung cancer(NSCLC) and the tumor cells of the regional metastasized lymph nodes, to explore the expression of CXCR4 in different clinicopathological characteristics, and further to explore the role of CXCR4 in regional lymph nodes metastasis in NSCLC.Methods: S-P immunohistochemical staining was used to investigate the expression of CXCR4 detected by rabbit anti-human CXCR4 monoclonal antibody. The specimens were 41 cases of primary tumor tissues of NSCLC, 25 cases of regional metastasized lymph nodes from those 41 NSCLC patients having regional lymph nodes metastasis, as well as 20 cases of normal lung tissues from the patients having non lung disease.
