商场室内氡暴露与外周血淋巴细胞遗传损伤的相关性研究

【摘要】 目的:通过氡暴露监测和人外周血淋巴细胞核DNA长度、微核发生率的检测,根据商场营业员职业氡暴露的累积剂量,探讨氡暴露与人体外周血淋巴细胞遗传损伤的相关性。 方法:在商场室内一楼和地下室连续24小时测量氡浓度。对40名研究对象,采用胞质分裂阻断微核法(CB微核法)检测微核细胞,用单细胞凝胶电泳技术(SCGE)测量DNA损伤情况。 结果:商场平均氡浓度结果分别为34.10±9.42 Bq/m~3,营业员氡暴露的年总有效剂量为1.0709mSv。在此暴露剂量下,人外周血淋巴细胞微核发生率为1.475‰,DNA彗星长度为26.46μm。累计剂量和年龄与微核细胞发生率之间的相关系数r分别为0.378 (p=0.016)、0.373(p=0.018),与核DNA长度之间的相关系数 r分别为r为0.172(p=0.08)、0.372(p=0.018)。 结论:1、本次调查的商场室内氡浓度未超过国家及地区标准。2、随着氡暴露累计剂量的上升,人体外周血淋巴细胞微核率和彗星长度显著增加,提示长期的氡暴露可导致淋巴细胞的遗传损伤。

【Abstract】 Objective: To estimete the effects of radon exposure on genetic damage of emporium workers by measurement of DNA length and micronucleus rate of peripheral blood lymphocytes.Methods: The indoor radon concentration was measured continuously 24 hours a day. The cytokinesis-block micronucleus and the single cell gel electrophoresis technique were applied to 40 objects to reflect the DNA damage by long term radon exposure.Results: The average radon concentration inside the emporium was 34.10+9.42 Bq/m~3 with the total effective dose per year of 1.0709 mSv. The average micronucleus rate was 1.475‰ and the kernel DNA length was 26.46 μ m. The correlation coefficients between accumulative dose with the age and the micronucleus rate were 0.378 (p=0.016) and 0.373 (p=0.018) , and those with the age and the DNA length were 0.172 (p=0.08) and 0.372(p=0.018 ) , respectively.Conclusions: The emporium indoor radon concentration was under the National and State standands. With the accumulative dose of radon exposure, the micronucleus rate of peripheral blood lymphocytes and the DNA length increased correlatively, indicating that long-term radon exposure could reduce
