两种植物多糖的分离纯化、结构鉴定及初步生物学活性的研究

Study on Isolation, Purification, Structure Identification and Elementary Biological Activities of Polysaccharides from Two Kinds of Plant

【摘要】 本文以土人参和鸡矢藤为研究对象,材料经乙醇脱脂、热水浸提、Sevage法脱蛋白、乙醇沉淀等处理得粗多糖;粗多糖以DEAE-纤维素离子交换柱层析、SepharoseCL-6B凝胶柱层析等方法进行纯化得单一多糖成分;然后用凝胶柱层析和聚丙烯酰胺凝胶盘状电泳鉴定纯度,凝胶渗透色谱(GPC)法测定分子量,气相色谱、红外光谱、紫外光谱、纸层析等技术初步分析了其结构;并对单一多糖成分的相关生物学活性进行了研究。最终从这两种植物中获得五种单一多糖成分,而这五种单一多糖成分及其相关活性,目前未见相关研究报道,具体研究结果如下。从土人参中分离出两种单一多糖:TPP3a和TPP5b。TPP3a为酸性多糖,其糖含量为79.04%,糖醛酸含量为22.66%;数均分子量Mn为5.275×104,重均分子量Mw为1.076×105;由鼠李糖、阿拉伯糖和半乳糖组成,其摩尔比为:0.81:1.04:1.00;主要含有吡喃糖,糖苷键构型主要为β型。TPP5b为杂多糖,其糖含量为73.74%;数均分子量Mn为7.682×103,重均分子量Mw为8.452×103,由甘露糖和葡萄糖组成,其摩尔比为:1.00:4.50;主要含有呋喃糖。采用诱导PC12细胞分化模型,以神经生长因子(NGF)为阳性对照,在20μg/mL浓度下,TPP5b对PC12细胞的生长有一定的促分化作用,TPP3a无相关活性。从鸡矢藤中分离出三种单一多糖:PSP2a、PSP2b和PSP3a。PSP2a为酸性多糖,其糖含量为69.12%,糖醛酸含量为3.62%;由阿拉伯糖、半乳糖和葡萄糖组成,其摩尔比为:1.00:0.92:1.41;主要含有呋喃糖。PSP2b为酸性多糖,其糖含量为85.33%,糖醛酸含量为7.38%;由阿拉伯糖、半乳糖和葡萄糖组成,其摩尔比为:1.00:0.73:1.80;主要含有呋喃糖。PSP3a为酸性多糖,其糖含量为64.43%,糖醛酸含量为25.57%,由鼠李糖、阿拉伯糖和半乳糖组成,其摩尔比为:1.23:1.00:0.55;主要含有吡喃糖,糖苷键构型主要为β型。采用感染前腹腔给药方式,以链霉素为阳性对照,鸡矢藤粗多糖PSP(1000mg/kg/d),鸡矢藤多糖组分PSP2(1000mg/kg/d),鸡矢藤单一多糖成分PSP2a(500mg/kg/d)对受铜绿假单胞耐药菌感染的小鼠均有不同程度保护作用。小鼠存活率分别达到10%,30%,40%,以PSP2a的效果最好。多糖的活性因多糖纯度的提高而增强。

【Abstract】 Talinum paniculatum Gaertn. and Paederia scandens (Lour.) Merr. were separately extracted by ethanol and hot water successively. The water extract were deprotinized by Sevage method, precipitated by ethanol, and then the crude polysaccharides were obtained. The crude polysaccharides were isolated and purified by DEAE-cellulose ion-exchange column chromatography and Sepharose CL-6B gel column chromatography, and then the homogeneous polysaccharides were gained. The purity of homogeneous polysaccharide was identified by gel column chromatography and polyacrylamide disc gel electrophoresis, and molecular weight was identified by GPC. Their structure was analyzed by GC, IR, UV and PC. The biological activities of homogeneous polysaccharide were elementarily investigated. Finally, five kinds of homogeneous polysaccharides were obtained from the two kinds of plant. So far, the five kinds of homogeneous polysaccharides and their biological activities have not been reported yet, and the related results as follows.Two kinds of homogeneous polysaccharides, TPP3a and TPP5b, were obtained from Talinum paniculatum Gaertn.. TPP3a was an acidic polysaccharide, consisted of rhamnose, arabinose and galactose in the molar ratio of 0.81:1.04:1.00. Its saccharide and uronic acid content were 79.04% and 22.66% respectively. TPP3a mainly contained pyranose and linked byβ-glycosidic bond in its backbone. The number-average molecular weight (Mn) and weight-average molecular weight (Mw) of TPP3a were 5.275×104, 1.076×105 respectively. TPP5b was a neutral heteropolysaccharides, consisted of mannose and glucose in the molar ratio of 1.00:4.50. Its number-average molecular weight (Mn) and weight-average molecular weight (Mw) were 7.682×103, 8.452×103 respectively. TPP5b mainly contained furanose and its saccharide content was 73.74%.Under the model of induction of PC12 cell differentiation, NGF as positive control, TPP5b (20μg/mL) could induct PC12 cell differentiation in vitro, but TPP3a couldn’t. Three kinds of homogeneous polysaccharides, PSP2a, PSP2b and PSP3a, were obtained from Paederia scandens (Lour.) Merr.. PSP2a, an acidic polysaccharide, mainly contained furanose and consisted of arabinose, galactose and glucose in the molar ratio of 0.81:1.04:1.00.
