肾炎防衰液对慢肾衰大鼠的疗效观察

【摘要】 本研究采用5/6肾切除方法制造慢性肾衰竭大鼠模型,通过观察肾炎防衰液对肾衰竭大鼠体重、肾重与体重之比、血压、24小时尿量、24小时尿蛋白平均浓度、血生化指标(血肌酐、尿素氮)以及病理染色(HE染色、Mallory染色、六胺银染色)的改变,研究其对慢性肾衰竭的防治作用。 1.实验目的 观察肾炎防衰液对5/6肾切除导致的慢性肾衰竭大鼠的防治作用。 2.实验材料与方法 2.1 实验动物 SD雄性大鼠(北京维通利华实验动物技术有限公司提供)体重:180~200g,健康。 2.2 实验试剂和药物 尿毒清(广州康臣药业有限公司提供),洛汀新(北京诺华制药有限公司,批号:05048),肾炎防衰液(北京中医药大学药系提供)。 2.3 模型制作与分组用药 SD雄性大鼠,90只,先随机分为三组,正常组9只、假手术组9只、手术组共72只。然后实行假手术和手术,适应性喂养一周后,手术组大鼠再随机分为6组,分别是:模型组、尿毒清阳性对照组、洛汀新阳性药物对照组、肾炎防衰液小剂量组,肾炎防衰液中剂量组,肾炎防衰液大剂量组。正常组和假手术组每天给予3ml生理盐水灌胃,治疗组每天上午给予相应药物灌胃。药物预防治疗的时间为15周。 2.4 标本的收集与指标的检测 实验过程中,密切观察大鼠的行为、体征等改变。在大鼠灌胃前、灌胃第2周后、第6周等后分别称量大鼠的体重;灌胃第4周时、第6周、第8周、第12周、第15周时分别留取每组大鼠的24小时尿量并测定尿中的平均蛋白浓;测量灌胃4、8、12、15周时,大鼠尾动脉血压。灌胃第15周后留取大鼠的血样、肾组织并且给肾组织称重,血液经低温离心后分离血清,存放待测。7150自动生化分析仪测定血样中的肌酐、尿素氮、血脂(胆固醇、甘油三酯、低密度脂蛋白)。用SPSS11.5系统进行数据的分析,数据用(?)±S表示,结果采用方差分析。

【Abstract】 Studying the function of preventing and curing of Shenyan Fangshuaiye on the chronic kidney failure through building the chronic kidney failure model of rats caused by 5/6SNX, and observing the weight of rats, the weight ratio of kidney and body, Blood pressure, 24-hour urine amount, 24-hour urine albumen , and biochemical index(blood serum creatinine, Blood urea nitrogen), and pathology stainings(H-E staining, Mallory staining ,amine silver staining).1. PurposeObserve Shenyan Fangshuaiye’ s prevention and cure function on chronic kidney failure of rats that 5/6 SNX caused.2. Experiment material and methods2.1 Experiment animalSD male rats (offered by Beijng Weitonglihua Experiment animal technique limited company), weight: 180~200g, health.2.2 Experiment reagent and medicineNiaoduqing(offered by Guangzhoukangcheng department biotechnology technique limited company), Luodingxin(Beijing Nuohua department biotechnology technique limited company, authentication code: 05048), Shenyanfangshuaiye ( offered by the Chinese herbal medicine college, Beijing University of Chinese Medicine).2.3 Model building, group and treatFirstly, 90 SD male rats were divided into 3 groups at random, :normal group(9), false operation group(9) and operation group(72). Then operate and adaptability feed them for two weeks. Operation group mice were divided into 6 groups at random, including: Model group, the control group of Niaoduqing, the control group of Luodingxin , the low dose group of Shenyan Fanfshuaiye, the median dose group of Shenyan Fanfshuaiye, the heavy dose group of Shenyan Fangshuaiye. All the rats belong to normal group and false operation group were given 3 ml Physiology brine aevery day. While the others were given homologous drugs every morning. All the treatments lasted 15 weeks.
