细胞体外定量缺氧培养实验测控系统的研制

【摘要】 探究缺氧状况下人体细胞的发生、发展、结构与功能、病变机制、衰老死亡的特征已逐渐成为生物医学研究的一个活跃领域。而构建缺氧培养环境作为确立细胞缺氧损伤模型实验的基础工作是极其重要的。但由于生物体本身是复杂的自组织自适应系统,在体实验往往受到体液、呼吸、循环等复杂因素的影响,很难解释某单一因素的影响作用和强度。所以建立具有一定精确度的细胞体外定量缺氧培养实验装置,成为目前生物医学工程研究领域研究的热点。 本论文基于细胞体外缺氧培养的实验要求,开展了定量缺氧培养测控系统的研制工作,并根据实验结果作了相应的校正优化。论文研究的主要内容及特色如下: (1) 分析国内外相关文献资料,总结现有的构建缺氧培养环境的实验方法,以CO2培养箱为对象提出了以计算机技术为核心开发细胞体外定量缺氧培养测控系统的设计思路。 (2) 基于硬件控制,研制了细胞体外定量缺氧实验装置。并从抗干扰的角度,采用迟滞电压比较电路,改进了氧浓度控制电路。避免了系统在实验过程中电磁阀所出现的频繁开闭现象。 (3) 基于计算机控制,开发细胞体外定量缺氧实验测控系统。以实现细胞体外定量缺氧培养为基本目标,选择高精度氧变送器,配合多功能数据采集卡,完成整套系统的数据采集工作。再结合相应的数据处理算法,利用阈值比较实现对执行机构——电磁阀的通断控制。并且从测控操作的实际需要出发,构建测控软件的操作界面,不仅使系统的调控工作更加灵活,也实现了对监控环境中氧浓度的实时观测。

【Abstract】 Exploring the cell’s occurrence, development, configuration and function, mechanism of pathological changes, characteristic of consenescence and decease has being the active field in the domain of biomedical research. And As the basic for the whole experimental work, designing the model of hypoxia cell culture is very important. However, organism is a complex system which organizes and accommodates both by itself, so owing to the staggering complexity of the in vivo environment, establishing an experimental system for hypoxia cell culture in vitro proved with acceptable precision is the indispensable experimental method in the biomedical researches.An experimental system for hypoxia cell culture in vitro supported was established in this paper, on the elementary demands of the experiment for hypoxia cell culture. The following results were attained,(1) The thesis reviewed the current literature and summarized the up-to-data experimental devise and methods for hypoxia cell cultured. The merits and demerits of each design were especially analyzed.(2) A devise for hypoxia cell culture was established based on the hardware control design.(3) A measuring and controlling system for hypoxia cell culture established based on the PC control. The modified system can not only process the sampling data more efficiently with the data pretreatment and fixed value control arithmetic, butalso provide an interface for manipulating and observing in real time.(4) Measuring and controlling system Emendation by experimentation considering the sampling delay for the gas diffusing. And acquire the delay coefficient used in optimized arithmetic. After emendation by experimentation, the precision reaches 0.8% in the oxygen consistence controlling range froml% to 5%, and reaches 0.7% above 5%.(5) Aiming at the deficiency of on-off control system, this paper provided a optimized arithmetic, PID Control with the Smith Precompensation, from the continuous control theory. Theoretic preparative was made for further improvement.
