复合物理场对微生物的诱变作用

【摘要】 本论文对本实验室筛选得到的一株产纤维素酶细菌LBT-3.6进行了紫外、微波复合诱变,选育到一株滤纸酶活力提高了202.7%的突变株WB-2。同时进行了发酵条件的优化,最佳条件为:麸皮50g/L,(NH43PO4 0.4%,蛋白胨0.1%,NaCl 0.4%,Tween800.2%,接种量3%(V/V),pH8.0,30℃培养48h,滤纸酶活力达到12.35U/mL。还研究了红、黄、蓝、绿和紫色五种发光二极管对WB-2生长的影响。 同时对生物表面活性剂产生菌L-21进行了紫外诱变,筛选到一株糖脂产量提高了112.5%的突变株LZ25.11。

【Abstract】 Microbial breeding selection with physical mutagens was investigated in this paper. The strain WB-2 of high yield cellulase was the mutant of strain LBT-3.6 treated by UV and microwave, its filter paper activity was increased by 202.7%. The optimum parameters for fermentation consisted of 50g/L wheat bran, 0.4% (NH4)3PO4, 0.1% peptone, 0.4% NaCl, 0.2% Tween80, pH8.0. The maximum FPA of WB-2 reached 12.35U/mL after growth at 30癈 for 48h on a rotary flask shaker. Effect on growth of WB-2 was studied using five LEDs, which contained red, yellow, blue, green and purple.The strain LZ25-11 of high yield biosurfactant was obtained after UV treatment from L-21, its output of glycolipid was increased by 112.5%.
