沙棘果实中黄酮类成分的提取、分离与鉴定

【摘要】 国外对沙棘黄酮的研究较为深入和广泛,已分离鉴定出32种黄酮化合物,但国内对沙棘黄酮研究的报道较少。本文对蒙古沙棘果实中黄酮类化合物的提取、分离和鉴定进行了研究。 本研究是用先用甲醇冷浸、石油醚脱脂、乙酸乙酯萃取得到提取物,聚酰胺柱层析结合高效液相色谱进行单体化合物的分离,但提取时溶剂比例(5:5:3:3:1)、分离时聚酰胺粉目数(80-100目)和上样量(常规的1/4)与传统有所差别,从蒙古沙棘的果实中分离得到了3个黄酮单体化合物,本实验操作流程简单,适合于工业化生产。通过物理常数、化学方法和波谱分析[紫外(UV)、质谱(MS)、核磁共振(NMR包括1H-NMR、13C-NMR)]鉴定出其中3个黄酮单体化合物(化合物Ⅰ,Ⅱ,Ⅲ),化合物Ⅰ的母环为槲皮素,化合物Ⅱ,Ⅲ的母环均为柽柳黄素。化合物Ⅰ被鉴定为槲皮素-3-芸香苷(Quecertin 3-O-rutinose),化合物Ⅱ鉴定为柽柳素-3-O-葡萄糖苷(Tamarixetin 3-O-glueoside),化合物Ⅲ鉴定为柽柳素-3-O-β-D-葡萄糖-7-O-α-L-鼠李糖苷(Tamarixetin 3-O-β-D-glucoside 7-O-α-L-rhamnoside)。化合物Ⅱ、Ⅲ为首次从沙棘属植物中分离得到,经CA检索认为是新化合物。 黄酮类化合物广泛分布于植物界,沙棘中含有丰富的黄酮化合物,沙棘黄酮是沙棘的主要活性成分,具有降低胆固醇、抗氧化、抗衰老、抗溃疡、抗炎和抗癌等生理功效及显著的药用功能。本研究为提取、分离、和鉴定沙棘果实中所含黄酮类化合物提供了科学方法,并为深入开发和利用沙棘资源,尤其是对沙棘黄酮类药物的产业化生产提供了新方法。

【Abstract】 Extraction and isolation and identification of flavones in seabuckthorn (Hippophae rhamnoides L.) fruits were studied in this paper. Flavones of Hippophae rhamnoides have been at abroad , 32 flavones extracted and identified, but there were a few reports studies on Flavones of Hippophae rhamnoides at home, so it is necessary to extensive study about extraction, isolation and identification of flavones.Three compounds were isolated from the fruits of seabuckthorn with the extraction of organic (MeOH 5: 5: 3: 3: 1) solvent and with of isolation of polyamide chromatogramphic column (80-100) and high performance liquid chromatogramphy. On the basis of physical constants, chemical evidence and spectrum analysis[UV, MS, NMR (including 1H-NMR, 13C-NMR) ] , compound I basic structure is Quecertin, compounds II, III basic structure are Tamarixetin.three compounds (I, II, III) were identified. They are Quecertin 3-O-rutinose(I), Tamarixetin 3-O-glucoside(II)and Tamarixetin 3-0- P -D-glucoside 7-O- a -L-rhamnoside(III) Compounds II, III were identified from Hippophae .L for the first time and confirmed as new compounds by searching Chemistry Abstract (CA) .Three new flavones compounds were isolated from the fruits of(Hippophae rhamnoides L. subsp. Mongolia Rousi). This provides scientific basis for flavones compounds spectrum identification.Flavones compounds are extensive existence in the plants , Flavones of Hippophae rhamnoides have been regarded as the principal components responsible for the pharmacological activity of Hippophae rhamnoides fruits, Flavones of Hippophae rhamnoides have been studied in relation to a wide range of physiological functions such as inhibition of low density lipoprotein oxidation, hypercholesterolemia, antioxidant, antidecrepitude, antiulcer, antivirus , anticancer and so on by the many reports on the pharmacological, chemical, biochemical studies. This study provides a scientific basis for extracting, isolating, purificating, identifying on flavones of Hippophae rhamnoides h., in order to further to exploite and utilize seabuckthorn resource, especially provides new method for Flavones of Hippophae rhamnoides medicine industrialization production .
