中药色谱指纹图谱研究

【摘要】 本论文采用二极管阵列检测器和电化学检测器,研究了中药活性成分含量测定的高效液相色谱方法及其指纹图谱。论文中所取得的研究成果对于中药材质量控制有重要意义,也为确定中药材GAP生产基地提供了依据。全文共分五章,作者的主要贡献有以下四个方面: 1、建立了不同品种、不同采收部位和不同采收时间绞股蓝HPLC指纹图谱,实验结果显示:不同采收部位、不同采收时间、不同品种绞股蓝药材的色谱图有较明显的差异,各成份含量亦不同。本研究为快速鉴别区分不同品种的药材提供科学的依据。 2、采用高效液相色谱法建立了测定天麻中主要活性成份天麻素等含量的方法,建立了天麻的高效液相色谱指纹图谱。实验结果表明:不同产地的天麻中主要活性成份含量有明显差别,但其含有的整体化学成分无明显差异。该法为天麻药材的鉴定提供了较全面的信息。 3、采用高效液相色谱法建立了测定喜树不同部位中喜树碱含量的方法,并通过对不同产地、不同部位和不同生长期喜树中的主要活性成份相对保留时间的计算获得其色谱指纹图谱。实验结果表明:不同产地喜树种子、不同部位喜树叶子和不同生长期喜树茎部的色谱指纹图谱中峰个数及形状有明显差别,其中叶子和茎部都有其特征色谱峰;且三者的喜树碱含量差异较大。该研究为我国喜树及喜树碱的产业化提供理论基础依据。 4、采用高效液相色谱电化学检测器,直接、连续测定了不同产地的葡萄酒中7种酚酸类化合物的含量,并建立了葡萄酒中酚酸类化合物的色谱指纹图谱。实验结果表明:7种酚酸得到很好的分离,其浓度与峰面积之间的线性关系良好,并且电化学检测器的检测灵敏度是光检测器的4~600倍,该研究成果为葡萄酒的质量控制提供了科学依据。

【Abstract】 In this thesis, high performance liquid chromatography (HPLC) was applied to determine the contents of active components and to establish its chromatographic fingerprint in Traditional Chinese Medicine (TCM) by electrochemical detector (ECD) and diode array detector (DAD). Those studies are very important to control the quality of TCM, and can offer a basis for the Good Agricultural Practice (GAP) management. This thesis consisted of five chapters, and the author’s contributions are in the following four aspects:1. The chromatographic fingerprint of gynostemma pentaphyllum makino in different season and its different part was established by HPLC. The experimental results showed that the chromatograms of gynostemma pentaphyllum makino were obviously different in all samples, and the content is different in all samples. This investigation can provide a basis for the quality control of gynostemma pentaphyllum makino.2. A new method for determination of the contents of active components in gastrodia elata blume was presented by HPLC, and an ideal fingerprint of gastrodia elata blume was established. Experimental results showed that the number and shape of peaks on chromatograms of the gastrodia elata blume obtained from some different resource were basically same, but content of gastrodin as a main component was obviously different in those samples. This investigation may provide some useful information for identification of gastrodia elata blume and a basis for the quality control of gastrodia elata blume. It is very important to management of GAP and toexploitation and use of TCM.3. A new method for determination of the contents of active components in camptotheca acuminate obtained form different resources was presented by HPLC, and an ideal fingerprint of camptotheca acuminata was established by calculation of the relative retention time and similarity of main active ingredients on the chromatograms of camptotheca acuminata. Experimental results showed that the number and shape of peaks on HPLC fingerprint of the camptotheca acuminata were different, but content of camptothecin as a main component was obviously different in the different samples; the contents in leafage were higher than that in seeds, which was superior to stems. This method will be useful in the future human studies with the industrialization of camptotheca acuminata and camptothecin.4. To establish an HPLC-ECD method for determination of seven kinds of phenolic compounds in five different wine samples simultaneously, and an ideal fingerprint of wine was established by HPLC-ECD. The experimental results showed that seven kinds of phenolic compounds are separated satisfactorily and there are some good linear relationship between their concentration and peak area. When ECD was applied, the sensitivity of phenolic compounds is 4~600 times than that obtained with UV detection. The study may provide a scientific basis for the quality control and research of wine.
