壳聚糖中药复合药膜对皮肤溃疡大鼠EGF、PDGF影响的实验研究

【摘要】 慢性皮肤溃疡(Chronic skin ulcer,CSU),是一种常见的难治性疾病,发病率高,治疗困难,中医药在治疗上有其独特的优势。本实验从分子水平出发,研究壳聚糖中药复合药膜治疗慢性皮肤溃疡的机理。 本实验通过试管法研究壳聚糖中药复合药膜对金黄色葡萄球菌、绿脓杆菌、大肠杆菌、变形杆菌、表皮葡萄球菌、肺炎克雷伯杆菌的抑制作用。以Wistar大鼠为研究对象,依据文献复制了大鼠背部皮肤溃疡模型,以贝复剂为对照药物,观察治疗前后皮肤溃疡的愈合情况,通过双抗夹心ELISA法检测治疗前后皮肤溃疡大鼠血清EGF、PDGF的表达水平。结果显示:壳聚糖中药复合药膜对于各种细菌均有不同程度抑制作用。对变形杆菌的抑制作用最好,其次是绿脓杆菌,对金黄色葡萄球菌、大肠杆菌、表皮葡萄球菌、肺炎克雷伯杆菌也有不同程度的抑制作用。造模成功后,大鼠血清EGF明显降低,PDGF明显升高。经治疗后,各组血清EGF、PDGF呈抛物线变化,但都在造模后的基础上有不同程度的升高,达到峰值后降低,但仍高于正常水平。壳聚糖中药复合药膜治疗组的变化尤为明显,达到峰值的时间提前,血清中浓度增高,与空白组、贝复剂组比较,有统计学意义,明显优于各组(P<0.01)。本实验提示:壳聚糖中药复合药膜对各种细菌有较强的抑制作用;壳聚糖中药复合药膜可以提高血清EGF、PDGF的表达水平,提前达峰时间,从而促进皮肤溃疡的愈合。

【Abstract】 Chronic skin ulcer is a kind of popular disease, including vascular ulcer, pressure ulcer, radioactive ulcer and infectious ulcer, etc. The incidence of the disease has increased considerably in the past few years, but the cure rate was not satisfactoried. Traditional medicine treating the disease has individual advantage. The research has studied the basic mechanism of Chitosan Chinese-medicine multiple membrane treating chronic skin ulcer on the molecular level.This experiment study has researched the anti-bacteria activity of staphylococcus aureus , colibacillus , Pseudomonas aeruginosa , Klebsiella pneumoniae, Bacillus proteus, staphylococcus epidermidis of Chitosan Chinese-medicine multiple membrane by test-tube method. Back ulcer models were established on rats with literature. Then model rats were divided into three groups and treated by Chitosan Chinese-medicine multiple membrane, Physiology and Beifuji respectively. Adopted the tissues of ulcer surface separately and observe their growth circumstance by light microscope and electric microscope . Observe the variation of Epidermal growth factor , Platelet derived growth factor before and after therapy by double-anti-ELISA. The result showed that Chitosan Chinese-medicine multiple membrane has a certain of antibacterial action . The best antibacterial action in Bacillus proteus and the second is Pseudomonas aeruginosa. After model made successfully , Epidermal growth factor dropped while Platelet derived growth factor increased. The result suggested that Chitosan Chinese-medicine multiple membrane group changes obviously, reach the climax before the due date. These were different obviously compared with the other two groups (P<0.01) .The research shows Chitosan Chinese-medicine multiple membrane has great antibacterial action. It can cure the chronic skin ulcer by increasing the growth of EOF and PDGF, reach the climax before the due date.
