人工腱膜材料组织相容性的初步研究

【摘要】 目的:对人工腱膜材料的组织相容性进行初步研究,了解其生物特性。 方法:1)动物埋植试验:将人工腱膜、动物的自体腱膜以及同种异体腱膜分别植入大白兔体内,于手术后4、8、15周取出,观察其局部组织学变化的有关指标,对三种埋植物引起的局部反应进行比较。2)热原反应试验:将人工腱膜浸取液通过耳缘静脉注入大白兔体内,分别于注射后1h、2h、3h测量兔的肛温与注射前的正常肛温进行比较。3)全身过敏试验:将人工腱膜浸取液0.5ml,共3次,肌肉注射健康豚鼠体内,然后将其随机分成两组,分别于首次注射后的第14天、第21天经腹腔注入人工腱膜浸取液2ml,观察过敏症状与阳性对照组比较。4)眼结膜刺激试验:选大白兔6只,分别于动物右眼滴入人工腱膜浸取液2滴,左眼滴入等量2%柠檬酸液,经1~3d观察眼结膜有无充血、水肿和局部组织反应。 结果:1)动物埋植试验显示:人工腱膜在埋植后8周,周围组织炎症反应消退;在埋植后15周,人工腱膜组织形态与自体腱膜组织形态在光镜下观察无差异,成纤维细胞计数及淋巴细胞计数,人工腱膜与自体腱膜、同种异体腱膜比较,在统计学上无显著差异(P>0.05)。2)热原反应试验:人工腱膜组织浸取液注入动物体内,无致热反应。3)全身过敏试验:人工腱膜组织浸取液注入动物体内,无全身过敏反应。4)眼结膜刺激试验:人工腱膜组织浸取液对家兔眼结膜无刺激性。 结论:人工腱膜材料对机体无刺激性,植入体内无致热反应及全身过敏反应,在植入动物体内15周,其形态结构及组织相容性与自体腱膜无差异,该材料具有良好的组织相容性,是临床治疗腱膜组织缺损的理想修补材料。

【Abstract】 OBJECTIVE:To analyse the biocompatibility of artificial fascia preliminarily.METHOD: l)eighteen rabbits were implanted with artificial fascia ,autograft and isograft, the local histological reaction was observed in different time.2 ) Six rabbits were injected with the extract of artificial fascia ,the temperature was observed in different time.3)Six guinea pigs were injected with the extract of artificial fascia and then they were divided into two groups .One group were injected with the extract of artificial fascia again 14 days later and another group were injected again 21 days later .We observed the reaction of the guinea pigs and the result was compared to the control group.4)The extract of artificial fascia was dropped in the eyes of rabbits ,the reaction was observed in different time.RESULT: l)There was no different about the local histological reactionamong the three groups after 15 weeks.2) The temperature of the rabbits didn’t become higher than before. 3) The guinea pigs had no allergic reaction after they were injected with the extract of artificial fascia .4)The extract of artificial fascia was no irritant to eyes.CONCLUSION:The artificial fascia is biocompatible and suitable as graft material.
