女性肝郁状态下神经递质、激素的实验研究

【摘要】 目的：复制肝郁证动物模型，并予经轻胶囊干预，探讨妇女肝郁状态下神经递质的变化及其引起的生殖内分泌的改变，并初步分析肝郁导致女性情感性疾病的机理及中药经轻胶囊防治其发生发展的机制。 方法：用绷带束缚大鼠四肢，外加束缚桶进行造模，以经轻胶囊为实验用药，对照观察大鼠一般情况、精神、行为、体重的变化；用荧光分光光度法检测大鼠中枢单胺类神经递质NE、DA、5-HT的变化；用放射免疫法检测大鼠血清E₂、P、PRL、T的变化及下丘脑β-EP含量的变化；用免疫组化法检测下丘脑ER的改变。 结果：肝郁造模组大鼠一般情况、精神、行为、体重发生了明显改变；中枢单胺类神经递质NE、DA的含量及5-HT/DA的值异常升高；血清E₂、P分泌增多，E₂/P的比值异常升高；下丘脑β-EP及ER含量亦异常升高。造模加中药组大鼠一般情况、精神、行为、体重得到明显好转；中枢单胺类神经递质NE、DA及5-HT/DA得以调节；血清E₂、P分泌异常得到缓解；下丘脑β-EP及ER含量也得到明显改善。 结论：用捆绑法复制的肝郁证动物模型，是一个比较理想的中医证候动物模型。肝郁可致雌性大鼠中枢单胺类神经递质合成代谢异常及雌激素分泌紊乱，从而导致一系列精神情志改变和生殖紊乱。疏肝理气的经轻胶囊能改善肝郁模型实验大鼠的症状及体征，对单胺类神经递质、内分泌具有明显调节作用。更多还原

【Abstract】 Objiective: Producing depression of liver-Qi animal models and giving Chinese medicime-JingQing Capsule to interfere to study the change of neurotransmitter and breeding endocrine. Moreover to make a preliminary analysis for the reason of depression of Iiver-Qi leading to Female affective psychosis and for the mechanism of JingQing Capsule in preventing and curing female depression of Iiver-Qi.Method: The animal model was made by means of binding up rat’s limbs and putting it into special moulds. Using JingQing Capsule as trial drug to smooth the circulation of liver-Qi. We observed the change of rat’s general condition, spirit, behavior weight. Detecting the change of NE, DA, 5-HT in the rat brain stem by fluorospectiophotometry and the change of E2, P, T, PRL in the rats serum and B -endorphin in hypothalamus by radioimmunoassay. Detecting the content of ER in hypothalamus by immunohi stochemi cal method.Result: Some changes of the general condition spirit, behavior and weight of model rats occurred;The content of neurotransmitter abnormally rised up such as NE, DA, 5-HT and E2, P in the rats serum also rised up;ER and B-endorphin in hypothalamus both stepped up obviously. However above-mentioned targets appeared differential reversion in rats of the treated groop.Conclusion: The animal model of Iiver-Qi depression made by means of binding up is an ideal TCM animal model. Liver-Qi depression can lead to abnormality of neurotransmitter and estrogens So to cause diseases of psyche and genital system.更多还原
