苦参碱生物学特性研究

【摘要】 本研究采用高效毛细管电泳法测定苦参碱的含量,证明方法的重现性、精确性均达到要求,得出此方法较常规的HPLC法简便、快速、经济,具有较高的实用价值。同时,在此基础上采用改进寇氏法测定了大鼠口服苦参碱的半数致死量即LD50为542.13±72.12mg/kg,95%的可信限为617.93~473.69mg/kg。采用打孔法进一步测定了苦参碱对大肠杆菌,绿脓杆菌,金黄色葡萄球菌及链球菌的敏感程度。采用倍比稀释法测定了苦参碱对上述四种菌的最低抑菌浓度,分别是0.0156mg/ml,0.0313mg/ml,0.0156mg/ml,0.0625mg/ml。本实验还通过对实验和靶动物制造大肠杆菌病的病理模型,测定出苦参碱在100mg/kg的剂量下对大肠杆菌所致雏鸡死亡率有显著的降低作用。说明苦参碱对大肠杆菌有很好的抑制作用,为进一步了解苦参碱的特性,开发和利用苦参碱奠定了基础。

【Abstract】 This study introduced the new method of high performance capillary electrophoresis (HPCE) to measure the content of matrine. The result shows that this method measures up to the requirements in recurrence and accuracy, more convenient, faster and more economical than general method of HPLC, and is a practical method. At the same time, based on the above result, the LD50 of rats oral matrine was measured by the improved Karber method. The result shows that the LDso is 542.13±72.12mg/kg, and the 95% of acceptable range is 617.93~473.69 mg/kg. In addition, this study measured the sensitivity degree of matrine to Escherchia coli, Pseudomonas aeruginosa S.aureus and Streptococcus, by using the stiletto method. The minimum concentration to t restrain the above four kinds of bacteria is 0.0156mg/ml , 0.0313mg/ml, 0.0156mg/ml and 0.0625mg/ml. respectively. By using the experiment animal and target animal to establish the pathology model of Colibacillosis, the experiment also measured out that matrine can effectively reduce the death rate of chicken caused by the Escherchia coli at the dose of 100mg/kg, showing that matrine has a good prohibiting effect on the Escherchia coli. The above results will help to learning more about the property of matrine, and to the exploitation of matrine.
