灵芝与红曲霉的高密度发酵及开发应用研究

【摘要】 灵芝与红曲霉均含有多种对人体有益的生物活性物质,已经受到了微生物学工作者和医药工作者人的关注,并进行了初步的开发研究。为了拓展这两种真菌的生物活性产品种类及提高其生产水平,我们对灵芝和红曲霉进行了较为深入的研究。 在灵芝的研究中,我们进行了灵芝的培养基和培养条件的优化试验,并在此基础上进行了深层液态分批发酵、补料分批发酵,使灵芝的生物量达到了9.29g/L,灵芝多糖的产量在60h时达到了2.208g/L,合成速率为0.036g.h-1.L-1,灵芝多糖的合成速率比文献报道的增加了20%。在发酵的过群中我们还发现,灵芝多糖的合成是非生长关联型的,而是开始于稳定期。根据发酵试验的实际情况,若经过代谢调控延长稳定期,有可能还会进一步提高灵芝多糖的产量。这些实验结果预示着通过液态发酵的方式进行灵芝多糖的工业化生产完全是可行的。 我们还进行了灵芝多糖的下游加工工艺条件的研究,确定了灵芝多糖的提取工艺和蜂胶中的黄酮的提取工艺,并以灵芝多糖和黄酮为君药,配以黄芪、丹参和元参,开发出了蜂参灵胶囊制剂,该制剂具有增强人体免疫功能和降低血糖之功效。 在红曲霉的研究中,我们进行了红曲霉的培养基和培养条件的优化实验,并根据该实验结果进行了高密度发酵,最终红曲霉菌丝体的含量达到了4.09g/L,比我们所看到的2.18g/L高出近一倍。但是,由于种种原因,关于Monacolin类物质在发酵过程中的动态变化目前我们无法监测,这部分的工作只好留待日后进行了。 本论文进行的灵芝与红曲霉发酵条件、动力学参数和代谢调控的研究为其工业化生产提供了重要的技术支持。此外,利用灵芝发酵产物研制出的蜂参灵胶囊为糖尿病患者提供了一种新的药品。

【Abstract】 Both Ganoderma lucidum and Monascus purpureus contain many bioactive substances which are healthful to humans. Lots of microbiologists and medical workers have paid much attention to these microorganisms and made some progress in primary studies. In order to increase the products yields from G lucidum and M. purpureus and improve their production level, these fungus were studied deeply in this paper.For the research of G lucidum, a series of experimnents were carried out to find out the optimum medium compositions and the optimum culitivation conditions. By using liquid fermentation method, batch and fed-batch were carried out. Finally, 9.29g/L of biomass of G lucidum was obtained and 2.208g/L of its polysaccharide was attain after 60 hour-fermentation. The synthesis rate of the polysaccharides was enhanced to 0.036g .h-1.L-1 which was 20 % higher than the data reported. In addition, we found that the synthesis method of G lucidum polysaccharides was unrelatable type with growth, and the polysaccharides was synthesized at the stable phase. According to the actual situation of the synthesis process, the yield of the polysaccharides would be improved if the stable phase had been prolonged by adjusting the metabolism artificially. These results indicate that the industrialized production of G lucidum polysaccharides by liquid fermentation is completely feasible.The downstream processing conditions of G lucidum polysaccharides and flavone were studied and the extracting technique routes of these were set up. With the G lucidum polysaccharides and flavone as the "monarch drug", Feng Shen Ling Capsules have been developed with radix astragali, radix salviae miltiorrhizae and radix scrophulariae as the "principal drug". The capsule has been proved effective in enhancing human immunity function and decreasing the blood sugar level.In the study of M. purpureus, the optimum conditions for its medium composition and culitivation were discussed. High density fermentation was carried out and the mycelium yield reached to 4.09g/L, which is about two times higher than the data of 2.18g/L reported in literature. However, the dynamic parameters of the Monacolin substances couldn’t be investigated in the process of fermentation due to some reasons.In this paper, fermentation conditions of G. lucidum and M. purpureu were studied respectively in their dynamic parameters and metabolism regulation. This will provide important technological supports for further industrialized production. Furthermore, Feng Shen Ling Capsula developed from the fermentation products of G lucidum will provied a new promising medicine for curing diabetes.
