百合中秋水仙碱的分离应用研究

【摘要】 秋水仙碱是1820年于百合科植物秋水仙中发现的一种重要的卓酚酮类生物碱。由于秋水仙碱及其类似物的特殊生理活性和药用价值,它们一直受到广泛的关注。 论文选取我省湘西龙山县的百合为研究对象,并从中提取了有效活性成分—秋水仙碱。 本文对天然植物中秋水仙碱的研究进展作了详细的综述,给出了秋水仙碱的植物资源、秋水仙碱的合成、秋水仙碱的结构、性质、鉴定方法、功能及应用;着重研究了超临界二氧化碳流体萃取百合中的秋水仙碱的工艺条件,并用正交法对工艺条件进行了优化筛选,找到了超临界二氧化碳流体萃取百合中秋水仙碱的最佳工艺条件:在40℃、18MPa压力条件下,以乙醇为提携剂萃取2h。同时将各因素对萃取效果影响的大小进行了比较,在选定的工艺条件内,各因素的影响秩序为:萃取温度>萃取时间>萃取压力>提携剂用量。同时,本文用传统的有机溶剂提取法对百合中的秋水仙碱进行了提取研究,并与超临界流体萃取方法进行了比较。结果表明:超临界流体萃取秋水仙碱步骤简单、条件温和,一般超临界萃取设备均可达到这些条件,易于实现工业化。超临界流体萃取不但萃取效率高,产生的“三废”少,对环境友好;而且超临界萃取物中杂质成分较少,便于后期的分离纯化。因此,此法可应用于秋水仙碱的提取。 论文通过高效液相色谱法对超临界萃取物中秋水仙碱含量进行了分析。测得百合中秋水仙碱的含量为0.049%。并用薄层色谱法对萃取物进行分离纯化,采用~1HNMR、IR法对目标物的结构进行了表征。 论文还将百合提取液对羟自由基的清除作用进行了研究,从而为开发百合复方保健品提供理论依据。

【Abstract】 Colchicine, which is a principal tropholone alkaloid, was isolated from Colchicum autumnale Liliacease in 1 820. Much attention was paid to colchicine and its analogues for their special activities and officinal value.In this paper, an physiological component colchicines was studied in Bulb of Lily ,which was taken from longshan county , Hunan province. Researches on colchicine from the Bulbs of Lily abroad were reviewed, including resources of colchicine, methods of extraction, methods of separation and characterization.Extraction of colchicine from the bulbs of lily was studied by supercritical carbon dioxide fluid and the extraction technological conditions were optimized by vertical-method. In selected technological conditions, the influence order of each factor is: extraction temprature > extraction time > extraction pressure > pre-soaker dosages.The content of colchicine in extracts was determined directly by high performance liquid chromatography (HPLC). The results showed that at 40 , 18MPa and with ethanol as pre-soaker and extract 2h.. Under the given conditions, colchicine concentration increased greatly from 0.049% in natural sample to 6.4% in extract with supercritical fluid extraction(SFE). The comparison of SFE and organic solvent extraction suggested that theformer is a quick, safe, non-toxic and environmental friendly method with higher yield and selectivity. So supercritical fluid extraction of colchicine from the bulbs of lily is practicable.Pure colchicine was isolation from the extracts through thin-layer chromatography and it was identified by using spectral techniques such as1HNMR, IR.Meanwhile, Rearch on effect of lilium extract for scavenging hydroxyl radical(OH), It can be applied to develop healing ointments of lily.
