不同剂量清热解毒化浊片对大鼠免疫性肝损伤的实验研究

【摘要】 目的:探讨清热解毒化浊片对大鼠免疫性肝损伤的作用机理,并确定用药的最佳剂量方法:采用尾静脉攻击注射牛血清白蛋白(BSA)造成大鼠免疫性肝损伤模型。将受试大鼠随机分成空白对照组、模型组、清热解毒化浊片小剂量组、大剂量组,然后分别施以相应处理,从肝功能、肝纤维化血清学指标、肝组织病理学方面观察不同剂量清热解毒化浊片护肝、抗肝纤的作用,并探讨其最佳剂量。同时,观察其CH50、CIC、TXA2-PGI2系统及氧化/抗氧化指标的影响,以阐述该药的作用机理。结果:1.大、小剂量清热解毒化浊片均可显著降低ALT、CG,与模型组比较P<0.01。大剂量清热解毒化浊片还能显著升高TPR、ALB及PCⅢ水平,与小剂量组比较P<0.05。2.清热解毒化浊片大、小剂量组大鼠肝细胞炎症活动度及肝纤维化程度均较模型组轻,清热解毒化浊片大剂量组较小剂量组又更为轻微。3.大、小剂量清热解毒化浊片均能降低血浆CIC水平,与正常组比较P>0.05,升高血浆CH50水平,与模型组比较P<0.01。4.大、小剂量清热解毒化浊片均能降低血浆TXB2水平,纠正T/K比值,与模型组比较,P<0.01,大剂量清热解毒化浊片还能降低MDA、升高血浆6-Keto-PGF1α水平,与模型组比较,P<0.01,与小剂量组比较,P<0.01。结论:大、小剂量清热解毒化浊片均能有效改善肝脏功能,保护受损肝细胞,并能抗肝纤维化,但大剂量清热解毒化浊片疗效较小剂量佳,其机理与清除MDA、CIC,抑制CIC激活补体及纠正TXA2-PGI2系统失衡有关

【Abstract】 This animal experiment is to probe into the effect and mechanism of Tablet Qingrejieduhuazhuo on rats given BSA to induce immunity liver injury by observing rat’s liver pathological change,ALT、HA、PCⅢ、CG、CH50、CIC、MDA、TXB2、6-Keto-PGF1αand so on,and to find out the best dosage of it.The results showed:1. Tablet Qingrejieduhuazhuo can markedly lower ALT、CG,yet,the big dosage of Tablet Qingrejieduhuazhuo can increase the level of TPR、ALB and PCⅢ in blood. 2. The big dosage of Tablet Qingrejieduhuazhuo have better effect on improve liver cell inflammation and hepatic fibrosis than the little dosage.<WP=6>3. Tablet Qingrejieduhuazhuo can lower the level of CIC,and increase the level of CH50 in blood.4. Tablet Qingrejieduhuazhuo can lower the level of TXB2,rectify the ratio of T/K,and the big dosage of Tablet Qingrejieduhuazhuo can decrease the level of MDA、increase the level of 6-Keto-PGF1α.Conclusions: Tablet Qingrejieduhuazhuo have pretty effect on protecting hepatocyte and reject hepatic fibrosis,and the effect of the big dosage is better than the little dosage.The mechanism might have anything to do with eliminating MDA、CIC,restraining CIC’s activiating the complements and revising the dysequilibrium of TXA2-PGI2
