MyoG、Myf-5基因在申农1号品系猪中的多态性及其与肉质相关性的研究

【摘要】 本文采用PCR-RFLP分析方法,对33头申农Ⅰ号品系育肥猪进行了MyoG基因和Myf-5基因的研究,分析了各基因型之间在初生重、断奶重、系水力、剪切力、肌内脂肪及瘦肉率等性状间的差异。从而找出MyoG基因和Myf-5基因与生长性状和肉质性状之间的相关性,尝试为猪的育种工作提供分子标记。 研究结果表明,MyoG基因和Myf-5基因都存在多态性。MyoG基因用PCR1 Msp Ⅰ-RFLP法检测,表现为两种基因型,AA型和AB型;MyoG基因用PCR1 Msp Ⅰ-RFLP法检测,表现为三种基因型,MM型、MN型和NN型;Myf-5基因用PCR-Hinf Ⅰ-RFLP法检测,表现为两种基因型,EE型和EF型。通过SAS分析,表明申农Ⅰ号品系猪的MyoG基因的PCR-Msp Ⅰ-RFLP位点与猪的初生重、系水力相关(p<0.05),与断奶重、剪切力及肌内脂的不相关(p>0.05)。申农Ⅰ号品系猪的Myf-5基因的PCR-Hinf Ⅰ-RFLP位点与猪的初生重、肌内脂相关(p<0.05)。

【Abstract】 Polymorphisms of MyoG gene and Myf-5 gene in Shennong No.I pig line were studied in the experiment, by means of PCR-RFLP. The differences of birth weight, weaning weight, water holding capacity, shear force, intratnusclar fat and carcass lean percentage among different genotypes and the polymorphisms of MyoG gene and Myf-5 gene were analysed. The correlation bewteen the two genes and growth performance with meat quality would be found out to identify a molecular marker for pig breeding.The results showed that there were polymorphisms of MyoG gene and myf-5 gene in Shennong No.I pig line. The genotypes of MyoG gene PCR1 Msp I-RFLP have AA and AB; MyoG gene PCR2 Msp I -RFLP have MM, MN and NN, and Myf-5 gene PCR Hinf I -RFLP have EE and EF. Polymorphisms of MyoG gene were correlated with birth weight and water holding capacity in Shennong No.I pig (p<0.05), and polymorphisms of Myf-5 gene were correlated with birth weight and intramuscular fat(p<0.05).
