用SAP,DXA及骨生物力学测定观察复方中药对去卵巢大鼠骨质疏松模型的疗效

【摘要】 本文目的:研究复方中药对去卵巢大鼠骨密度的影响。方法:采用去势大鼠骨质疏松模型,将59只3月龄雌性大鼠随机分为假手术组(与模型组实施同样的手术但不切除卵巢)、模型组、尼尔雌醇组、复方中药(淫羊藿、黄芪、女贞子等)小剂量组、中剂量组和大剂量组。用单光子吸收法(SAP)测定各组大鼠左侧股骨远侧干骺端骨矿含量(BMC)、骨密度 (BMD)。用双能X-线骨密度测量仪(DXA)测量左侧股骨骨密度(BMD)。用电子万能试验机测定大鼠右侧股骨最大载荷(E)和破坏挠度(R)。结果 SAP测得模型组的BMC及BMD与假手术比较差异显著(p<0.01),与阳性对照组及小、中、大三个剂量比较差异显著(p<0.01)。DXA测得模型组与假手术组的BMD差异显著,复方小剂量骨密度较模型组有上升趋势,其他用药各组骨密度与模型组相比无明显差异;复方中剂量组与模型组相比最大载荷及破坏挠度值有明显上升(p<0.01),差异显著,复方中剂量药物对改善骨质疏松后的骨的生物力学性能有作用。结论:复方中药有提高OP模型的骨密度的作用,在中剂量时能提高去卵巢骨质疏松模型骨生物力学性能,增加骨组织的承载能力,提高骨骼抵抗外力冲击能力。说明复方中药在治疗去势大鼠骨质疏松症中,疗效是肯定的。

【Abstract】 Objective:To study the effects of NF(new fomula) on the bone mineral dencity (BMD),ultimate load (UL), ultimate deformation (UD) of ovariectomized rats.Methods fifty-nine 3-month-old femal SD rats were randomly divided into six groups:false operation group(all were operated without spay); ovariectomized Group (OVX);estrin group;NF(new formula)groups (epimedium,astyagalusroot,gloss privet fruit,etc) with low dosage(NF+L) ;NF groups with moderate dosage(NF+M); NF groups with high dosage(NF+H);The experiment last for three months .At the end of it ,the BMD and BMC of left femoral distial metaphysis of all groups was determined with SAP;The BMD of left femur was measured with DXA. Ultimate load (UL) and ultimate deformation (UD) of right femur of all groups were measured with electronic universal experimentai apparatus..Results; The BMC and BMD of rats’ left distal femur of OVX group which were measurd with SAP was significantly lower than that of false operation group (p<0.01), the BMC and BMD of other groups were significantly higherer than that of OVX group (p<0.01).Tthe BMD of rats’ left femur of OVX group which were measurd with DXA was significantly lower than that of false operation group (p<0.05); BMD of NL+L group showed an increasing trend compared with OVX group.BMD of other groups had no significane of difference as compared with OVX group .The UD and UL of NF+M group was significantly highter than that of OVX group(p<0.01). NF+M played a role in improve biomechanics nature of bone with osteoporsis.Conclution :New formula improve the BMD and BMC of the OVX rats. NF+M may improve the bone biochemical properties of OVX-rats;The bearing capacity of bone tissue was increased;The resistance of skeleton against external force was improved. The results suggsested that the :new formula capsule improve the bone loss of ovariectomized rats and prevent osteoporosis.
