马尾松毛虫质型多角体病毒全基因组序列的测定与分析

【摘要】 本论文由两章组成。第一章对质型多角体病毒的研究作了综述性介绍,包括对质型多角体病毒的分类、多角体及病毒粒子、复制及组装、基因组结构和功能等方面作了阐述。另外,本章还对马尾松毛虫质型多角体病毒的研究作了重点的介绍。第二章进行了马尾松毛虫质型多角体病毒(DpCPV)全基因组序列的测定与分析研究。马尾松毛虫质型多角体病毒属于呼肠孤病毒科、质型多角体病毒属,基因组由十条分段的dsRNA组成。在本文中,我们完成了DpCPV全基因组序列的测定工作。对测得的序列分析表明,每一基因组片段含一个开放性阅读框,两末端存在保守的核苷酸序列,分别为5’(AGUAA)和3’(GUUAGCC)。根据DpCPV-Hn编码的蛋白和呼肠孤病毒科其它成员编码的蛋白的比较结果推测,DpCPV-Hn S1、S3、S4、S6、S7分别编码病毒的结构蛋白VP1、VP2、VP3、VP4、VP5,S2编码蛋白为病毒的依赖于RNA的RNA聚合酶(RdRp),S5、S8、S9编码蛋白为病毒的非结构蛋白,S10为多角体蛋白基因。功能性结构域分析显示,VP3 氨基酸序列部分区域与Methanosarcina mazei Goel 的甲基化酶有同源性,推测VP3在病毒mRNA的加帽过程中起重要作用。VP4含有亮氨酸锌指结构和ATP/GTP结合位点,推测VP4具有核酸结合活性。S5编码蛋白的氨基酸序列部分区域与口蹄疫病毒的2A蛋白酶( FMDV 2Apro)非常相似。对几种CPV的RNA聚合酶基因和多角体蛋白基因的进化分析表明,DpCPV-Hn为I型CPV,和BmCPV的进化关系非常相近

【Abstract】 There are two chapters in this thesis.Chapter one is a general introduction of cytoplasmic polyhedrosis virus (CPV), including the classification about CPV, the polyhedrin and virus particle of CPV and gene structures and functions of CPV. The research on Dendrolimus punctatus CPV (DpCPV) also has been introduced in this chapter.Chapter two is about the sequence determination and analyse of complete genome segments of Dendrolimus punctatus cytoplasmic polyhedrosis virus (DpCPV). DpCPV belongs to genus Cypovirus in the family Reoviridae. Its genome consist ten dsRNA segments. Each segment posses a single open reading frame. Conserved motifs 5’(AGUAA) and 3’(GUUAGCC) were found at the ends of each segment. Comparison of proteins of DpCPV-Hn with those of other members in the family Reoviridae lead us suggest that S1, S3, S4, S6 and S7 encode viral structural protein VP1, VP2, VP3, VP4 and VP5, respectively. S5, S8 and S9 encode viral non-stuctural proteins. S2 encodes an RNA-dependent-RNA polymerase (RdRp). S10 encodes polyhedrin. Motif analysis shows that VP3 is similar to the methyltransferase of Methanosarcina mazei Goel, VP4 has motifs for leucine zipper and ATP/GTP-binding sites, and the protein encoded by DpCPV S5 is remarkably similar to foot-and-mouth disease virus 2A protease (FMDV 2Apro). Phylogenetic analysis of RdRps from nine viruses of the family Reoviridae and polyhedrin from several characterized cypoviruses indicate that DpCPV-Hn is a type 1 cypovirus, more related to Bombyx mori cypovirus (BmCPV) than to other cypovirus species
