某些雌激素和抗癌光敏剂的分析方法研究

【摘要】 药物是用于治疗、预防疾病或调节机体生理功能并具有质量标准、经政府有关部门批准的天然来源物质或化学合成品。全面控制药物的质量,对于保证人民用药的安全、合理、有效具有重要的意义。随着分析技术的发展,药物分析采用的方法也层出不穷。化学发光分析法作为一种灵敏、快速的分析方法,近年来在药物分析上的应用日趋广泛。而电化学检测法作为一种高灵敏度与高选择性的测定电活性物质的方法,在药物的分析上有其独特的优势。本论文利用这二种方法进行一些药物的检测方法研究,具体有以下几个方面的内容:第一章阐述了化学发光分析法的原理、特点,对于在药物分析上常用的化学发光体系及常用的技术进行了介绍,同时对于本论文的主要研究对象----雌激素的检测手段及研究现状作了比较系统的阐述。第二章研究了一些雌酚类药物的化学发光性质,建立了用鲁米诺-过氧化氢体系测定雌酚类药物的新方法。该法用于测定己烯雌酚、双烯雌酚、己烷雌酚等物质,检出限均可达10-8 mol/L这一数量级。将本法用于片剂中己烯雌酚的含量测定,结果令人满意。本章还应用一些光谱学的方法对体系的发光机理进行了初步的探讨。第三章研究了苯甲酸雌二醇经硫酸质子化后的产物增强Ce(IV)-SO32-体系化学发光的现象,建立了检测苯甲酸雌二醇的化学发光分析方法。苯甲酸雌二醇的浓度在1.00×10-6 ~2.50×10-5 mol/L 范围内与化学发光强度呈现良好的线性关系,检测限为8.98×10-7 mol/L。将本法应用于苯甲酸雌二醇模拟样品的测定,结果令人满意。本章通过动力学曲线及荧光光谱对化学发光的机理也作了一定的探讨。第四章研究了酞菁抗癌光敏剂二邻苯二甲酰亚胺甲基二磺酸基酞菁锌的化学发光性质,建立了Luminol-H2O2- ZnPcS2P2-CTAB流动注射化学发光法体系。ZnPcS2P2线性范围为2.50×10-7 ~1.00×10-8 mol/L ,检测限为3.58×10-9 mol/L。该法具有很高的灵敏度,为酞菁类药物的检测提供了一种新的方法。<WP=4>第五章采用了线性扫描伏安法(LSV)、循环伏安法(CV)等现代电化学分析方法系统研究了炔雌醇在玻碳电极上的电化学行为,并应用微分脉冲伏安法(DPV)对炔雌醇进行定量分析。炔雌醇线性范围为4.0×10-6~1.0×10-4mol/L, 检测限为3.76×10-6mol/L,将本法用于实际样品的测定,简便、快速,结果令人满意。

【Abstract】 The medicine is natural source material or synthesize chemistries hich is used for the disease treatment and prevention or regulate machine physiology function combine , have the quantity standard and was granted by government department. Control the quantity of the medicine completely, have the important meaning for the public using medicine safely, reasonably and effectively . Along with the analysis technical developing , the method that medicine analysis adoption also piles up one after another. Be a kind of sensitive、quickly analysis method , Chemiuminescence method is extensively used in medicine analysis in recent years .Electricity chemistry determination as a high sensitive and high selectivity method for measure electric- active material having its special advantage in the analysis of medicine. This thesis makes use of the two kinds of examination method to proceed some medicine studies. the contents idiographic as follow:Part I, explain the principle, characteristics of hemiuminescence analysis method, and introduce the Chemiuminescence system and echnique which were common used for medichine determination .At the same time for the main research object of this thesis----the hormone of estrogen , the systemic expatiation of the examination means and the present study circs were applied . Part II, Study on Chemiuminescence characteristics of some estrogen medicine , and establishing a new method to measure estrogen medicine use the luminol - H2O2 Chemiuminescence system .The method used for measure DES,DIES ,HXS medicine, the determination limit to all can amount to 10-8mol/L quantity class. Used this methods for determinating DES troche, the result turn up trumps . In this part some spectrum methods also been applied to study the Chemiuminescence<WP=6>mechanism of the system. Part III, A Chemiuminescence analysis method was established when study the phenomenon of EB after sulfuric acid protonize strengthing the Ce( IV)- SO32- system Chemiuminescence.The linear range is 1.00×10-6 ~2.50×10-5 mol/L, the correlation coefficient is 0.9997 and the detection limit is 8.98×10-7 mol/L. The method has been applied to determinate EB in sample and result give satisfaction. A dynamics curve and fluorescence spectrums for this system help to study the mechanism of this Chemiuminescence system .Part IV, Study on the ZnPcS2P2 character and establishing he Luminol- H2O2- ZnPcS2P2- CTAB Chemiuminescence system . the linear range is 2.50×10-7 ~1.00×10-8 mol/L,the correlation coefficient is 0.9976 and the detection limit is 3.6×10-9 mol/L。The method has relative selectivity and high sensitivity .and offer some experiment bases for determination of phthalazinedione medichine .Part V, In this article, Linear Sweep Voltammetry(LSV)、Cyclic Voltammetry(CV)、Differential Pulse Voltammetry(DPV) were adopted for systemic study the Ethinylestradiol ˊs electrochemistry action on glassy carbon electrode. Experiment show that the oxidiation of EE on glassy carbon electrode is uncontrary, EE has only little adsorptivity on glassy carbon electrode. DPV method is applied for quantitative analysis of EE, and the linear range is 4.0(10-6(1.0(10-4mol/L ,the correlation coefficient is 0.9995, and the detection limit is 3.76×10-6mol/L。The method is conveniency and rapidly, it was offer to determine EE in sample and the result is satisfying.
