栀子黄、栀子甙提纯的研究

【摘要】 栀子黄和栀子甙存在于植物栀子的成熟果实,在食品加工业、饮料制造业、饲料工业及医药工业方面都有着广泛的应用。目前,栀子黄的生产工艺复杂,产品的得率和色价低,而栀子甙一股随废液直接排放,造成了资源的浪费。 本文以栀子果实为原料研究了栀子黄、栀子甙浸取和提纯的分离过程,通过对栀子黄浸取因素的研究,得到较为合适的浸取剂、浸取温度、浸取方式。实验结果表明较合适的浸取条件是:以50%乙醇水溶液(v/v)作浸取剂、在常温下浸泡栀子粉末、浸取时间为24小时、液固比为14:1。 采用大孔吸附树脂柱层析分离天色产物栀子黄,有目的地筛选了一些吸附树脂用于分离栀子黄,通过对各种树脂的静态单位体积吸附量和选择性的研究,选择NKA大孔吸附树脂作为分离栀子黄的吸附剂。以NKA大孔吸附树脂为研究对象,探讨了树脂对栀子黄的静态吸附速度和吸附曲线、乙醇浓度对树脂吸附的影响、不同浓度和流速下的动态穿透曲线,得到NKA吸附栀子黄较为合适的条件。研究了吸附后吸附柱的杂质洗脱和产品洗脱问题,发现经过去离子水、稀碱液、稀乙醇洗杂质后再用85%乙醇水溶液(m/m)洗脱得到的栀子黄洗脱液,干燥后得到色价大于420的栀子黄产品。同时还对NKA树脂的应用稳定性进行了研究,发现NKA树脂应用10次后,性能没有明显的降低。 研究了1300大孔吸附树脂对天色产物栀子甙的柱层析分离效果,探讨了1300树脂对栀子甙的静态吸附速度和吸附曲线、pH值对树脂吸附的影响、不同流速下的动态穿透曲线,得到1300树脂吸附栀子甙较为合适的条件。研究了吸附后吸附柱的杂质洗脱和产品洗脱问题,发现经过少量去离子水、稀乙醇洗杂质后再用85%乙醇水溶液(m/m)洗脱得到的栀子甙纯度较高。最后得到的产品中栀子甙含量大于75%。

【Abstract】 Crocin and geniposide consist in the mature fruit of gardenia , having got the extensive application in food processing industry, beverage manufacturing industry, animal feed industry and medicine industry. At present, the production technology of crocin is complex with the low yield and the low colority . Usually the geniposide is general to be exhausted directly with the waste liquid, leading to the resources wasted.This paper studied every kind of factors that affects the yield of extraction of crocin , getting better leaching agent, leaching temperature and leaching method. The experimental results show that better leaching conditions are that the leaching agent is 50% ethanol aqueous solution( v/ v) under the room temperature , leaching time is 24 hours, the ratio of gardenia to agent is 14:1.In this paper .the separation result of the crocin with macroporous adsorption resins by column chromatography was studied. Some adsorption resins were chosen for the purpose of separating crocin. By the experiments on the static unit volume adsorption capacity and the selectivity of every kind of resins for crocin , NKA macropore adsorption resin was chosen as the adsorbent of the crocin. Regarding NKA macroporous adsorption resin as the research object, the experiments on the static adsorption rate of the resin for crocin, the adsorption curve of the resin for crocin, effect of ethanol concentration on the behavior of the resin adsorption, the dynamic leaking curve under the different concentration, the dynamic leaking curve under the different flow rate were made and better conditions of NKA absorbing crocin were got. After studying the wash of the impurity and the product on adsorption column , it was found that after washing the adsorption column by deionized water, sparse alkali liquor , sparse ethanoland, and then crocin was eluted by 85% ethanol aqueous solution( m/ m), the crocin solution was very high easy to be dried, after drying .the colority of crocin is more than 420.An investigation of stability indicated that after adsorption and desorption for ten times, the stability of NKA resin was still excellent.In this paper, the separation result of geniposide with 1300 macroporous adsorption resins by column chromatography was studied. Through the experiments on the static adsorption rate of the 1300 resin for geniposide, the adsorption curve of the 1300 resin for geniposide, the behavior of the resin adsorption in different pH and the dynamic leaking curve under the different flow rate ,better conditions of 1300 resin absorbing geniposide were got. After studying the wash of the impurity and the product on adsorption column , it was found that after washing the adsorption column by a little deionized water and sparse ethanoland, and then geniposide was eluted by 85% ethanol aqueous solution( m/ m).Aafter drying geniposide solution . the purity of geniposide is more than 75%.
