接骨木根皮化学成分及其复方制剂成型工艺的研究

【摘要】 接骨木(Sambucus williamsii Hance)又名公道老、扦扦活等,为忍冬科接骨木属落叶灌木或乔木。我国分布广泛,资源丰富,主产于东北、华北各地,全株入药,其性平,味甘苦,主要用于治疗骨折、跌打损伤等病症;另外,接骨木果油具抗癌、降血脂和抗衰老作用。 接骨木民间用药虽然由来已久,但其现代研究文献报道较少,尤其是化学成分方面。本文利用溶剂法和色谱法从接骨木的干燥根皮90%乙醇提取物的正丁醇和氯仿可溶部分中共分得6种单体化合物。通过化学方法和1H-NMR、13C-NMR和MS等光谱法鉴定出从氯仿可溶部分中分离出的两个化合物分别为β-谷甾醇(β-sitosterol)和胡萝卜苷(Daucosterine),而从正丁醇可溶部分中分离出的4种化合物中的两种分别为α-莫诺苷(α-morroniside)和Caryoptoside,Caryoptoside为首次在接骨木中获得。另外两种化合物的结构鉴定工作仍在进行中。 此外,本文对应用大孔吸附树脂法大量制备α-莫诺苷的方法进行了探索性研究。结果表明,该方法简便易行;并以α-莫诺苷为考察指标,对以接骨木根皮为主要原料的治疗骨折的接骨木复方进行含量测定,含量为5.8%,该方法灵敏、重现性好。为了控制接骨木复方胶囊的质量,本文对接骨木复方胶囊的制剂成型工艺进行了研究,筛选出了制备接骨木复方胶囊的最佳辅料为玉米淀粉。 本文研究结果将为接骨木的进一步研究奠定了坚实的理论基础,同时也为接骨木的质量控制、临床用药、剂型改型及新药开发提供了可靠的科学依据。

【Abstract】 Sambucus williamsii Hance is distributed widely and abundantly in China. The complete plant is medicinal. It is mainly used for the treatments of rheumatic arthritis,injury from fall and fracture etc. Its oil can reduce blood-fat and resist senility.Six compounds have been isolated from the Sambucus williamsii Hance by chemical and chromatographic methods, two compounds (β -sitosterol and Daucosterine) from 95% EtOH extract,four from 50% EtOH extract,two compounds ( α -morroniside and Caryoptoside) have been identified by 1H-NMR., 13C-NMR and MS among the four compounds, the other two ones continue to be identified.The author has found the method to abundantly produce α -morroniside by macroporous adsorption resin. The method is simple and feasible. Quantitative determination by HPLC has been proceeded in compound prescription of Sambucus williamsii Hance, using α -morroniside as the evaluating criteria. The content is 5.8%.The author has also studied the preparation process of compound prescription of Sambucus williamsii Hance.
