冬虫夏草液体发酵富集硒、锌、锗研究

【摘要】 利用冬虫夏草对微量元素具有较强富集功能的特性,采用液体培养的方式对硒、锌、锗进行了富集与转化研究,获得了富硒、锌、锗等微量元素的菌丝体。通过正交实验、单因素实验等方法确定了冬虫夏草最佳摇瓶培养基为:马铃薯20%,葡萄糖2%,酵母粉0.4%,KH2PO4 0.2% MgSO4 0.15%,接种量5%,最适pH值为6.5,最适温度为25℃,最佳转速为120r/min。;单元素富集和三元素富集实验对硒、锌、锗的富集特性进行了研究,结果表明适宜浓度的微量元素能够明显促进菌丝体的生长和氨基酸的合成,但会影响腺苷和多糖的合成,三种元素的富集亦存在一定的相互关系;对富硒、锌、锗虫草菌丝体多糖进行了初步分离纯化及性质研究,证明虫草多糖至少含有三种组分,由葡萄糖、半乳糖和甘露糖组成。结果表明,冬虫夏草能够有效实现对硒、锌、锗的同时富集与转化,并且适宜浓度的硒、锌、锗能够明显促进冬虫夏草菌丝体的生长。

【Abstract】 Cordyceps sinensis submerged fermentation for bioaccumulation of Se, Zn and Ge was studied in this paper according to its super ability of bioaccumulation of trace elements, and the mycelia enriched with Se, Zn and Ge were obtained.In this paper, the optimal fermenting conditions were worked out through orthogonal test and single factor test. The result of optimization is: potato 20%, glucose 2%, yeast extract 0.4%, KH2PO4 0.2%, MgSO4 0.15%,inoculation 5%,pH6.5, 120r/min, 25℃.The characters about the bioaccumulation of Zn, Se and Ge were investigated by single element and triple elements bioaccumulation, then the functional ingredients in the mycelia enriched with Zn, Se and Ge were analyzed. The extraction and purification of cordyceps polysacchride gave us its primary physical and chemical charactors. The result indicates that Cordyceps sinensis can accumulate Se, Zn and Ge simultaneously and efficiently, and these elements with optimal concentration can highly accelerate the mycelial growth.
