心肌康注射液中黄芪化学成分及其指纹图谱的初步研究

【摘要】 中药注射剂是中药给药途径的一个突破,具有剂量小,发挥作用快,疗效确切等优点。根据我国传统中医药理论,心肌康注射液由黄芪和丹参两味中药组成,治疗病毒性心肌炎。心肌康注射液采用超滤技术,大孔吸附树脂分离纯化技术等现代科技手段,结合化学提取理论将中药复方中的活性分子提取,精制而制成。本研究制备了三批注射液。 由于中药是含多种成分的复合体,采用单一或少数几个成分做指标不能全面反映中药的质量,因此对其中蒙古黄芪的化学成分进行了研究,分离并制备了两种黄酮苷类化合物,作为蒙古黄芪及其制剂指纹图谱研究的指标性成分,为黄芪的质量评价提供了依据。 以芒柄花苷和9,10-二甲氧基紫檀烷-3-O-β-D葡萄糖苷为标准品,初步建立了蒙古黄芪半成品及成品制剂HPLC指纹图谱,采用梯度洗脱技术,摸索了一个在280nm波长条件下较佳的乙腈-水洗脱条件。并在该条件下,对两化合物进行了含量测定。 本实验通过对正丁醇萃取部位进行硅胶柱层析,用氯仿-甲醇进行梯度洗脱,分离出两种单体化合物,经过1~H-NMR、(13)~C-NMR、MS谱测定和结构解析,确定其化学结构为芒柄花苷,9,10-二甲氧基紫檀烷-3-O-β-D葡萄糖苷;通过HPLC含量测定,纯度达到了定性、定量对照晶的要求。

【Abstract】 The injection of TCM is a new way in administration of TCM . Xinjikang injection consists of the effective ingredients of Radix Salvia and Radix Astragali according to the basic theory of traditional Chinese medicine. This injection is an effective recipe to cure viral myocarditis ( VMC) . The article adopted modern scientific means such as ultrafitration and macropore adsorbed resin method combined with chemical extracted theory, separated effective compounds. A new kind of modern injection was made. We have made out three batches of injections Astragali.TCM is a complex compound, which contains various ingredients, single or a few ingredients can not reflect the quality of TCM properly. The study on the chemical constituents of Radix was carried out .The study has separated two chemical components, and found up a preliminarily Fingerprint chromatography of Radix Astragali.With the content of onoin and 9,10-dimethoxy-pterocarpan-3-O- ?-D-glucoside as index, this experiment has set up a preliminarily Fingerprint chromatography of intermediate and finished product in Radix Astragali by HPLC. With this condition, the experiment has determined onoin and 9,10-dimethoxy-pterocarpan-3-O- ?-D-glucoside in the three batches of injections.Standard substance of onoin and 9,10-dimethoxy-pterocarpan-3-O- 0 -D-glucoside has also been made. The purity of control substance accords with the demands of the standard. The structure is identified by !H-NMR> 13C-NMR.
