针刺对自然更年期大鼠作用机理的实验研究

【摘要】 导师吴高媛教授立足中医基础理论，结合现代医学研究成果，提出肾虚肝郁，冲任虚损是女性更年期综合征的病因病机，临床上采用针刺治疗本病取得很好的疗效。据此我们拟定补肾疏肝，调补冲任的治疗大法，以11～15月龄雌性SD大鼠为自然更年期模型，以电针足三里、三阴交、关元及太冲为针刺处方，来探索针刺治疗更年期综合征的作用机制。在电针治疗30天后测定血清雌二醇（E₂）、促性腺激素（FSH、LH）和白细胞介素-2（IL-2）及下丘脑单胺类神经递质[去甲肾上腺素（NE）、5-羟色胺（5-HT）、5-羟吲哚乙酸（5-HIAA）]含量，并做卵巢和肾上腺的组织形态学观察。实验结果表明：电针可使针刺组大鼠下降的E₂水平显著回升（P＜0.05），异常升高的FSH、LH明显下降（P＜0.01～0.05）：电针可明显升高血清IL-2水平（P＜0.05），提高更年期大鼠的细胞免疫功能：还可使针刺组大鼠升高的下丘脑5-HT、5-HIAA含量明显下降（P＜0.05），使降低的NE含量回升（P＜0.01），使升高的5-HT／NE比值降至青年对照组水平（P＜0.01）。通过以上实验研究，得出以下结论：电针可以调节更年期大鼠生殖内分泌、提高免疫力、调节植物神经功能，对更年期衰退的神经内分泌免疫网络（NEI-N）起综合性调节作用，具有多途径的作用机制和整体调节的特点。更多还原

【Abstract】 On the basis of T C M theory and the result of modern medical research, professor Wu GAOyuan pointed out that "Kidney-asthenia and liver depression, impairment of chong and ren channels" is the main pathogenesis of C S. And she acquired a good curative effect treated with acupuncture. According to this principle, we set up a treatment principle, that is "nourish the kidney and soothe the liver, invigorate Chong and Ren channels". We selected "Zusanli","Sanyinjiao","Guanyuan", "Taichong" as the prescription, used SD female rats of 11-15 months old as the natural climacterum models, to investigate the therapeutic mechanism of climacteric syndrome treated with electroacupuncture. After 30 days, we observed the change of serum E2 level, sexual stimulating hormone (FSH and LH), serum IL-2 and hypothalamic monoaminic neurotransmitter (NE, DA, 5-HT and 5-HIAA) of them. We also made the morphological observation of their ovary and suprarenal. The research indicated that electroacupuncture can evidently decrease the level of sexual stimulating hormone (p< 0.01-0.05) and elevate the level of serum E2 level of model rats (p<0.05); It can increase the content of serum IL-2 of model rats (p<0.05), it also can reduce the amount of 5-HT and 5-HIAA in the hypothalamus, which were increased in model group (p<0.05), elevate the amount of NE in the hypothalamus of acupuncture group (p<0.01); and decrease the ratio of 5-HT to NE nearly to the level of the youth group (p< 0.01). By means of research, we can draw a conclusion that eletroacupuncture can regulate reproduction endocrine and the function of vegetative nerve, it also can improve immunity of climacterum rats. It may produce a synthetic effect on nerve-endocrine-immune network of them, having the feature of totally regulating through multiway.更多还原
