电针对胃肠粘膜内分泌—免疫网络的调控作用

【摘要】 针灸可以调节脏腑机能。足阳明胃经“属于胃,联络脾脏,主治胃肠病”。针刺足阳明胃经上的穴位对胃肠功能有良好的调节作用已被大量的临床和实验研究所证实。这些调节作用对胃肠道的损伤及某些疾患有着修复和治疗作用,但其确切机理尚不十分清楚。随着人们对胃肠粘膜免疫的认识和研究的深入,其相关研究已成为一个热点,但是针刺对胃肠粘膜免疫的调节作用研究较少。本实验采用同步实验的方法观察了电针刺激足阳明胃经上不同穴位引起的胃肠粘膜免疫系统相关指标的变化及内在联系,旨在探讨针刺对胃肠免疫功能调控作用及可能机制。 方法:将大鼠随机分为足三里组、上巨虚组、非经非穴组及非针刺组。同步观察电针后某些脑肠肽及胃肠粘膜免疫功能的变化。应用放射免疫法测定外周血及胃肠组织中的SP、VIP、SS、sIgA的含量;应用免疫组化染色法观察胃肠组织中SP、VIP及SS的表达情况;应用四甲基偶氮唑盐微量酶反应比色法检测IEL和LPL的增殖活力。 结果: 1、电针足阳明胃经上的穴位可以使外周血及胃、肠组织中的sIgA的含量升高,可以使胃肠道的免疫功能得到提高其中针刺 承口旱区大学羽士学位诀丈 一 足三里穴对其影响最为明显,提示足三里穴具有穴位特异性。 2、电针足阳明胃经上的足三里穴可以使IEL和LPL的增殖 活力增加。IEL、LPL又与* 共同增强胃肠道的免疫功能。 3、电针足阳明胃经上穴位可以使外周血及胃、肠组织中的 SP、VIP及SS的含量均有升高,且与胃肠道的免疫功能密切相关。 推测针刺后脑肠肽可能主要通过外周和胃肠局部作用两个途径去 调节胃肠道的免疫功能。 4、免疫组化照片结果显示,针刺足三里在胃肠组织中的SP、 VIP及SS的阳性表达较非针刺多。 结论: 电针胃经穴位对胃肠粘膜免疫功能有良好的调节作用,而且 同一经络上不同穴位作用效果不同,体现了某些穴位对脏腑有相 对的特异性;以胃经主穴足三里穴为基础,初步证明了针刺足三 里穴可以明显提高胃肠粘膜免疫功能,同时,脑肠肽、体液因素 及内分泌系统也参与了对胃肠粘膜免疫功能调节作用。并且这些 因素之间相互作用、相互影响,对胃肠神经内分泌兔疫网络进行 有效的调节。

【Abstract】 Acupuncture can regulate the visceeral fiinction.The stomach channel of foot-Yangming(St),part of the stomach and coordinating the viscera,can be used for the treatment of gastrointestinal diseases.Lots of clinical and experimeatal studies have proved that stimulating the acupuncture points of St produces good regulatory effect on gastrointestinal function,and cures gastrointestinal injuries and after conditions.However.its true mechanism still remains unclear.With feather investigation and studies,more and more attention is paid of the gastrointestinal mucosal immunity.But the research into acupuncture’s effect on gastrointestal mucosal immunity is reported less.So in this sudy,based on synchronous experimeat,we stimulate the acupuncture points of St and measure its corresponding biochemical levels of gastrointestal immune system in order to evaluate acupuncture’s effect on the gastrointestal mucosal immune system and its possible mechanism. Methods40 rats were randomly divided into 4 groups :Tsusanli group.Shangchusu group,non-acupoint group and non-acupuncture group.Then the changes of brain-gut peptide and gastrointestinal mucosal immunity were synchronously observed.Radiommunoassy was used to determine the levels of SP,VIP,SS,sIgA in peripheral blood and gastrointestal tissue.The immunohistochemistry staining to observing SP,VIP,SS expression in the gastrointestal tissue.The proliferatio activity of IEL and LPL were detected by mononuclear cell cytotoxicity assay. Results1, EA on acupoints of St increased levels of slgA in peripheral blood and gastrointestal tissue,and so gastrointestal immunological function wasimproved,especially when Tsusanli point stimulated. 2> EA on Tsusanli increased lEL’s and LPL’s prolifeation activates.Tongether with slgA.IEL and LPL could reinforce the gastrointestalimmunological function. 3, EA on acupoints of St increased the levels of SP.VIP.SS in peripheralblood and gastrointestal tissueAccording to our experinental resultsand related literaturre,brain-gut peptide after acupuncturre can regulategastriintestal immunologic function through the peripheral roate andgastrointestal local effect. ConclusionsEA on the points of St produced good regulatory effects and the effects aried with different poines,which meant some points had relative visceral speaificity;it was proved that EA on Tsusanli enhanced significantly gastrointestinal immunological function, and brain-gut peptide and endocrine system wre also involved in the regulatory effect,the above factors interact each other and regulate effectively the gastrointestinal endocrine immunological network.
