中华绒螯蟹及中国对虾几种生化指标变化的研究

【摘要】 甲壳动物没有特异性免疫,对进入体内异物最主要的防御方式是血淋巴的吞噬作用。在吞噬过程中伴随大量活性物质的产生与释放,其中最主要的是氧自由基。本论文通过NBT还原法、钼酸铵比色法、TBA比色法、平板法、磷酸苯二钠法、L-多巴法等6种方法对超氧化物歧化酶、过氧化氢酶、丙二醛、溶菌酶、酸性磷酸酶、碱性磷酸酶、酚氧化酶等7种活性物质进行研究。其结果如下: 在氧供应不足或吞噬细胞被激活等情况下,产生大量的自由基,其中最主要的是超氧阴离子自由基。当其超出体内抗氧化酶的清除能力时,引起脂质过氧化链式反应,导致生物膜及微循环严重损伤,甚至崩溃,自由基及其产物使多糖聚解,不饱和脂肪酸过氧化,蛋白交联,RNA、DNA断裂、突变等。超氧化物歧化酶能特异性的清除超氧阴离子自由基。 患上岸病的中华绒螯蟹幼蟹其体内的超氧化物歧化酶活性分别是:8.6206、7.0608及6.4524U/mg protein;正常中华绒螫蟹幼蟹体内的超氧化物歧化酶活性分别为:35.0746、36.0926及35.4975 U/mg protein。上岸病中华绒螯蟹幼蟹体内的超氧化物歧化酶极显著的低于对照组(P<O.01)。 患白斑病的中国对虾其体内的超氧化物歧化酶活性分别是:-0.2281、-0.3632及-0.3310U/mg protein;正常中国对虾体内的超氧化物歧化酶活性分别是:2.2449、2.2471及2.2460U/mg protein。白斑病中国对虾体内的超氧化物歧化酶活性极显著的低于对照组(P<O.01)。 患抖抖病的中华绒螯蟹其体内的超氧化物歧化酶活性分别是:9.3714、9.8065及10.0007U/mg protein;正常中华绒螯蟹体内的超氧化物歧化酶活性分别是:1.6488、1.6562及1.6653U/mg protein;患抖抖病的中华绒螯蟹体内的超氧化物歧化酶活性极显著的高于对照组(P<O.01)。 患上岸病的中华绒螯蟹幼蟹其体内的过氧化氢酶活性分别是:10.7855、12.8221及11.8378KU/mg protein;正常中华绒螯蟹幼蟹体内的过氧化氢酶活性分别为:29.1108、27.4594及28.1518KU/mg protein。上岸病中华绒螯蟹幼蟹体内的过氧化氢酶极显著的低于对照组(P<O.01)。 患白斑病的中国对虾其体内的过氧化氢酶活性分别是:8.0269、7.5373及7.5373KU/mg protein;正常中国对虾体内的过氧化氢酶活性分别是:12.8156、11.1229及11.5331KU/mg protein。白斑病中国对虾体内的过氧化氢酶活性极显著的低于对照组(P<0.01)。 患抖抖病的中华绒螫蟹其体内的过氧化氢酶活性分别是:1.5004、0 7317及0.3419KU/msprotein;正常中华绒螫蟹体内的过氧化氢酶活性分别是:0.7379、0.7229及 1.5079KU/mg protein;患抖抖病的中华绒螫蟹体内的过氧化氢酶活性对照组无显著差异(P>0.05)。 患上岸病的中华绒螫蟹幼蟹其体内的丙二醛含量分别是:0.7690、0.7666及0.8143 nL TEP/mgprotein;正常中华绒螫蟹幼蟹体内的丙二醛含量分别为:0.4567、0.4723及0.4334 nL TEP/mgprotein。上岸病中华绒螫蟹幼蟹体内的丙二醛含量极显著的高于对照组(P<0.of)。 患白斑病的中国对虾其体内的丙二醛含量分别是:0.8460、0.8700及 0.8500 nL TEP/mgprotein;正常中国对虾体内的丙二醛含量分别是:0.8680、口.8440及 0.8440 nL TEP/mg protein。白斑病中国对虾体内的丙二醛含量与对照组无显著差异(P>0.05)。 患抖抖病的中华绒鳌蟹其体内的丙二醛含量分别是:0.7380、07468及 0.7502 nL TEP/mgprotein;正常中华绒鳌蟹体内的丙二醛含量分别是:0.3873、0.3880及 0,3873 nL TEP/mgProtein;患抖抖病的中华绒赘蟹体内的丙=醛含量极显著的高于对照组(P<0.of)。 主要还研究了甲壳动物的溶菌酶,并对甲壳动物酸性水解酶及碱性水解酶的测定方法建立起来。其溶菌酶的测定结果如下。 患上岸病的中华绒鳌蟹幼蟹其体内的溶菌酶活性分别是:3.5351、4.3351及 4.3351卜g/mgprotein;正常中华绒赘蟹幼蟹体内的溶菌酶活性分别为:6.5138、5.3314及 6.5138 ug/mgprotein。上岸病中华绒螫蟹幼蟹体内的溶菌酶活性极显著的低于对照组(P<0.of)。 患白斑病的中国对虾其体内的溶菌酶活性分别是:0.2033、0.1903及 0.2162卜g/mg protein;正常中国牙刘吓体内的溶菌酶活性分别是:0.2141、0.2417及 0.2003卜g/。g protein。白斑病中国对虾体内的溶菌酶活性与对照组无显著差异(P>0.05)。 患抖抖病的中华绒螫蟹其体内的溶菌酶活性分别是:26.3M5、32.3024及 ZI.4856卜g/mgprotein;正常中华绒螫蟹体内的溶菌酶活性分别是:31.1097、31.1097及 25.3718卜g/mmorotein;患抖抖病的中华绒螫蟹体内的溶菌酶活性极显著的高于对照组(P<0.of)。 本论文已建立起甲壳动物酚氧化酶的测定方法。 本论文从新的角度研究了中华绒赘蟹幼蟹上岸病、中国对虾白斑病及中华绒螫蟹抖抖病的体内兔疫能力变化,给这些病的防治以新的启迪。

【Abstract】 Crustacea have not specific immunity, phagocytosis of haemolymph is the main defense mode to exogenous matter. In the process of phagocytosis , lots of activity matter produce and set free , the most major is oxygen free in them. This dissertation studies superoxide dismutase(SOD), catalase(CAT), malondiadehyde(MDA), lysozyme(LSZ), acid phosphatase(ACP), alkaline phosphatase(ALP), phenoloxidase(PO) etc 7 kinds of activity matter by NET reduction method, ammonium molybdate, TBA method, flat method, bisodium benzene phosphate, L-dopa etc 6 kinds of method. The following is results:Under situation of hypoxia or phagocyte activated, organism produce lots of free radicals, 02~ is the most important. When it surpasses the removal ability of antioxidation , it .causes chain reactions of peroxidation, resulting in serious damnification of organism membrance and microcycle, even collapse. Free radicals and product of their metabolism make polyamylose divide chains, unsaturated fat acid peroxide, protein tangle, RNA and DNA rupture and mutate. SOD can remove O2 specifically.The activity of SOD of juvenile Chinese mitten-handed crab of disembarkation syndrome is respectively 8.6206,7.0608 and 6.4524 U/mg protein; that in the control juvenile Chinese mitten-handed crab is respectively 35.0746,36.0926 and 35.4975 U/mg protein. SOD in the disease crabs is remarkably lower than that in the control crabs(P<0. 01).The activity of SOD of Chinese shrimp of white spot syndrome is respectively -0.2281, -0.3632 and -0.3310 U/mg protein; that in the control Chinese shrimp is respectively 2.2449,2.2471 and 2.2460 U/mg protein. SOD in the disease shrimps is remarkably lower than that in the control shrimps(P<0. 01).The activity of SOD of adult Chinese mitten-handed crab of pectoral appendages trembling disease is respectively 9.3714,9.8065 and 10.0007 U/mg protein-, that in the control adult Chinese mitten-handed crab is respectively 1. 6488, 1. 6562 and 1. 6653 U/mg protein. SOD in the disease crabs is remarkably higher than that in the control crabs(P<0. 01).The activity of CAT of juvenile Chinese mitten-handed crab of disembarkation syndrome is respectively 10. 7855, 12. 8221 and 11. 8378 KL’/mg protein-, that in the control juvenile Chinese mitten-handed crab is respectively 29. 1108,27.4594 and 28. 1518 KU/mg protein. CAT in the disease crabs is remarkably lower than that in the control crabs(P<0. 01).The activity of CAT of Chinese shrimp of white spot syndrome is respectively 8. 0269, 7. 5373 and 7. 5373 KU/mg protein; that in the control Chinese shrimp is respectively 12. 8156, 11. 1229 and 11. 5331 KU/mg protein. CAT in the disease shrimps is remarkably lower than that in the control shrimps(P<0. 01).The activity of CAT of adult Chinese mitten-handed crab of pectoral appendages trembling disease is respectively 1.5004,0. 7317 and 0. 3419 KU/mg protein; that in the control adult Chinese mitten-handed crab is respectively 0. 7379,0. 7229 and 1. 5079 KU/mg protein. CAT in the disease crabs have not remarkably distinction with that in the control crabs(P>0. 05).The content of MDA of juvenile Chinese mitten-handed crab of disembarkation syndrome is respectively 0. 7690, 0. 7666 and 0. 8143 nL TEP/mg protein; that in the control juvenile Chinese mitten-handed crab is respectively 0.4567,0.4723 and 0.4334 nL TEP/mg protein. MDA in the disease crabs is remarkably higher than that in the control crabs(P<0.01).The content of CAT of Chinese shrimps of white spot syndrome is respectively 0.8460,0.8700 and 0.8500 nL TEP/mg protein; that in the control Chinese shrimp is respectively 0. 8680, 0. 8440 and 0. 8440 nL TEP/mg protein. MDA in the disease shrimps have not remarkably distinction with that in the control shrimps(P>0.05).The content of MDA of adult Chinese mitten-handed crab of pectoral appendages trembling disease is respectively 0. 7380,0.7468 and 0.7502 nL TEP/mg protein-, that in the control adult Chinese mitten-handed crab is respectively 0. 3873, 0. 3880 and 0. 3873 nL TEP/mg protein.
