稀释液营养剂和抗菌素对常温保存布尔山羊精液品质和受胎效果的研究

【摘要】 本研究的主要目的是筛选理想的布尔山羊精液常温保存的稀释液配方,使布尔山羊精液在常温下保存时间达到60小时以上,仍有正常的受胎率。稀释液的作用是为精子存活提供养分和能量;提供缓冲剂,防止乳酸形成过程中PH值变化的危险;抑制细菌生长,扩大精液量,提高与配母羊数,充分发挥优秀种公羊的利用。本文是以稀释液营养剂和抗菌素对常温保存布尔山羊精液品质和受胎效果进行的研究。本试验共设有五个稀释液配方Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ,其中Ⅰ、Ⅱ、Ⅲ号稀释液配方是为筛选理想营养剂而设计的,分别以葡萄糖、果糖和葡萄:果糖=1∶1的混合物为营养剂;Ⅳ、Ⅴ号稀释液配方是为筛选理想抗菌素而设计的,均以葡萄糖为营养剂,Ⅳ号稀释液配方以青、链酶素作为抗菌素,Ⅴ号稀释液配方是以庆大酶素作为抗菌素。在该试验过程中,主要测定了用不同稀释液稀释的布尔山羊精液在保存不同时间后的精子顶体酶活力、精液中透明质酸酶活力、精子活力、存活率、顶体完整率和畸形率及受胎率等指标。将其中九组数据分别与它们所对应的受胎率进行了相关性分析,分析结果表明,精子活力与受胎率的相关系数r=0.711(n=9P<0.05),精子存活率与受胎率的相关系数r=0.84(n=9 P<0.01),精子顶体完整率与受胎率的相关系数r=0.52(n=9 P>0.05),精子顶体酶活力与受胎率的相关系数r=0.58(n=9 P>0.05),精液中透明质酸酶活力与受胎率有明显的负相关r=-0.87(n=9 P<0.01),所以精子存活率和精液中透明质酸酶活力可作为预测受胎率的理想指标;同时,对用不同稀释液稀释的布尔山羊精液在保存60h后的精子活力、精子存活率、精子顶体完整率和畸形率、精液中透明质酸酶活力、精子顶体酶活力和受胎率进行方差分析显著性检验,若差异显著再进行多重比较。分析结果表明:以葡萄糖:果糖=1:1 混合物作为稀释液营养剂的保存效果较以其中任何一种作为营养剂的保存效果好:以庆大霉素作为抗菌素的保存效果比以青、链酶素作为抗菌素的效果好,因此筛选出了在常温条件下保存布尔山羊精液有理想效果的稀释液配方*,其主要成份为:葡萄糖、果糖、柠檬酸钠、EDTA、庆大霉素。

【Abstract】 Study on the Effect of Boer Goat Spermatozas Quality and Conception Rate Diluted with Different Solution at Normal Atmospheric Temperature Abstract The main purpose of the experiment was to select ideal diluent ingredient for preserving Boer goat spermatozas under normal atmospheric temperature, making Boer Goat spermatozes have normal conception rate when it was stored for 60 hours.Five formulas of solutions were devised in this experiment aiming at selecting ideal nutrients and antibiotics.Indexes were measured in this experiment such as Acrosin actirity, hyaluronidase activity, sperm vitality,acrosome integrality, sperm abnormality and conception rate. By data processing, the following results were abtained.There was no significant correlation between acrosin activity and acrosome Integrality, so were between acrosin activity and sperm vitality(P>O.05), while there was significantly positive correlation between acrosin activity and acrosome integrality, R0. 86(PKO .05), Between hyaluronidase activity and acrosome Integrality was significant correlation(P<0.05). Between hyaluronidase activity and conception rate was significantly negative correlation R=-O.07(P<O.05); Between sperm vitality and conception rate was significant correlation(P<O.O1); Hyaluronidase activity and sperm vitality were ideal indexes to forecast conception rate. The diluent containing glucose and Fructose was superior to that containing any one of them ,and the diluent containing gentamicin was superior to that containing penicillin and streptomycin for preservation Boer goat spermatozas. No LII was the ideal diluent formula consisting of Glucose ,Fructose, Sodium citric acid ,EDTA, Gentamicin.
