广炎灵针剂提取工艺初步研究

【摘要】 中药在治疗疾病方面有着独特的优势,一是天然药物毒副作用小,二是多种成分协同入药,可以产生单一化学成分无法比拟的治疗效果。但是,中药的生产和研究与国际同行相比较为落后,为提高产品的市场竞争力,较为可行的方法是从已有的方药中寻找具有研究潜力的突破点,进行科学的研究,将产品提高到具备与国际同类产品竞争的水平上。 丁香叶即是一种很有研究价值的药物,以其为原料制成的炎立消胶囊抗菌效果经受住了临床的检验。但考虑到其有效成分的化学性质和临床用药的要求,拟制成粉针。 本文主要研究了丁香叶中含量较大的有效成分之一,丁香苦苷,以硅胶柱层析的方法从中分离得到大量纯品,并经分析其核磁共振和质谱等信息得到确证。同时分离得到3,4-二羟基苯乙醇和甘露醇。 将丁香苦苷进行体内抑菌实验,以庆大霉素为对照药物,观察其对接种金黄色葡萄球菌小白鼠的抑菌效果。结果表明两者作用相似。 以丁香苦苷含量为指标,初步探索了不同型号大孔树脂的分离纯化效果,结果被考察的三种类型的树脂均可以很好的吸附保留丁香苦苷,纯化效果以H103型较为理想。

【Abstract】 Compared with chemical drugs,TCM has its unique advantages on low side effect and uncomparable cure function. Butwith the outdated procedures and study methods, their competitive abilities in the international market is too poor. In order to change this situation, a practive idea is to find break through in the existing drugs with scientific method.Syringa oblata Lindl. is a very promising plant as an antibacterial. It?s leaves have been made into YanLixiao Capsules which have well stood up to the clinical and market test. But consideredabout its chemical quality and clinical requirement, it should be made into Powder Inject ion.By column chromatography with silica梘el three compounds were isolated from the AcOEt extraction of its leaves. On basis of chemical and spectroscopy analysis the structures of them were established respectivelyas Syringopicroside (1) Cyclopenta [c] pyran?梒arbox?ylic acid,l?~桪梘lucopyranosyloxy)條, 4a,5,6,7,7a梙e?xahydro?梞etry?梠xo? 2?4梙ydroxyphenyl)ethyl ester, [ls<la,4aa,7a,7aa)],D梞annitol(2) and 3,4梔ihydro?xyphenethyl alcohol (13).Parmacological experiment on the antibacterial ability of Syringopicroside was carried and the result showed that Syringopicroside has the same effect on staphylococcus aureus compared with gentamicin.The absorption and purification abilities on Syringoproside were primarily studied using three types of macroporous resin named D1OL H1O3 and HPD500. The result showed that HPD1O3 is the most optimum one.
