更年乐治疗更年期综合征作用机理的实验研究

【摘要】 目的：探讨补肾疏肝、调补冲任的中药复方更年乐治疗更年期综合征的作用机理。办法：运用11～15月龄雌性SD大鼠，按照SD大鼠自然更年期模型的要求，将其分为更年期模型组与治疗组，另选3月龄大鼠作为青年对照组，分别给予生理盐水和更年乐煎剂灌胃处理。检测指标采用高效液相色谱-电化学法测定下丘脑单胺类神经递质（NE、DA、5-HT、5-HIAA）；用放射免疫法测定雌激素（E₂）和促性腺激素（FSH、LH）以及白细胞介素-2（IL-2），并做卵巢和胸腺的组织形态学观察。结果：①调节植物神经功能：更年乐可使模型组下丘脑升高的5-HT和5-HIAA含量明显下降（P＜0.01～0.05），使降低的NE含量回升（P＜0.05），升高的5-HT/NE比值降至接近青年组水平（P＜0.01）。②调节生殖内分泌系统：更年乐煎剂可明显增加模型组卵巢重量指数（P＜0.05），缓解其萎缩程度；并使模型组下降的E₂水平回升（P＜0.05），异常升高的FSH、LH明显下降（P＜0.01～0.05）。③提高低下的免疫功能：更年乐煎剂可使模型组降低的血清IL-2水平明显升高（P＜0.05），胸腺萎缩程度有所缓解。结论：中药复方更年乐通过调节紊乱的植物神经功能、失调的生殖内分泌系统和低下的免疫功能，可能多层次、多途径、多环节地作用于更年期衰退的神经内分泌免疫网络，从而达到稳定机体内环境，缓解更年期症状的作用。更多还原

【Abstract】 AbstractObjectives:To study the therapeutic action on climacteric syndrome of Chinese herbal formula Geng-Nian-le (GNL), which can nourish the kidney, soothe the liver and invigorate Chong and Ken channels. Methods: According to the standard of nature climacterum model, we used l1-i5months old SD female rats as models. Meanwhile, we also selected 3 months old SD female rats as controls. Give GNL to some of the models, and give 0.9%NaCl solute to others. Hypothalamic monoaminic transmitters (NE, DA, 5-HT and 5-l-IIAA) contents were determined by high performance liquid chromatography and electrochemical detector. Serum E2, FSH, LU and IL-2 were detected by radioimmunoassay. Results:egulating the function of plant nerve: GNL can reduce hypothalamic 5-I-IT and 5-l-IIAA,which were increased in models (P < 0.0l-0.05) ;and it can increase reduced NE of hypothalamus in models (P < 0.05) ; also it can let the ratio of 5-HT to NE decrease nearly to the normal level group (P ?0.01) ;㏑egulating reproduction endocrine: GNL can evidently lighten ovary atrophy degree, improve serum E2 level and obviously lower serum FSI-I and LU levers of model rats.?Improving immunity: GNL can increase the content of serum lL-2 in models (P < 0.05) and lighten the atrophy degree of their thymus. Conclusions: By means of regulating thd function of plant nerve and reproduction endocrine and improving immunity, GNL may produce an effect on nerve-endocrine-immune network by various levels, ways and links. So it can stabilize inner condition of body and relieve symptoms of climacteric syndrome.更多还原
