中强度静磁场对肿瘤细胞生长的生物学效应研究

【摘要】 目的探讨中强度静磁场对鼠黑色素瘤细胞B16和人神经胶质瘤细胞U251生长的生物学效应,为磁场在肿瘤疾病的治疗中的应用提供理论依据。方法本实验分三个部分研究中强度静磁场对B16和U251生长的影响。(1)采用MTT法测定经50-200mT静磁场暴露48h和72h后的细胞存活率,检测静磁场暴露对细胞活力的影响。(2)利用流式细胞术测定静磁场暴露48h内的细胞周期分布情况,探讨静磁场影响肿瘤细胞生长的机制。(3)利用生物化学方法测定静磁场暴露48h内与细胞氧化防御系统相关的蛋白酶活性及抗氧化剂水平,检测静磁场对肿瘤细胞代谢过程的影响,探讨其与静磁场影响肿瘤细胞生长的机制的关联。结果(1)对B16和U251进行短时间50-200mT静磁场暴露,可以抑制细胞生长,但随着暴露时间延长,又转为促进作用。(2)100mT和200mT静磁场暴露对B16和U251的细胞周期分布没有影响。(3)100mT和200mT静磁场暴露48h后,B16和U251的GST活性和GSH/GSSG水平表现为先上升后下降;SOD活性和T-AOC能力先下降后上升;CAT活性基本没有受到影响。结论中强度静磁场可以抑制B16和U251的生长,诱导肿瘤细胞产生氧化应激。

【Abstract】 Objective: This research is to investigate the bioeffects of exposure tomoderate-intensity static magnetic fields on the growth of mouse melanoma cells Bl6and human glioma cells U251, and provide a theoretical basis for the application ofSMF in the treatment of tumor disease.Methods: The three sets of experiments were conducted in this study.(1) AfterB16and U251cells were exposed to moderate-intensity static magnetic fields(50-200mT) for48hours and72hours, cell growth was measured by MTT assay toexamine the effect on cells viability;(2) Flow cytometry was used to detect cell cycledistribution in48hours of exposure to static magnetic fields, to explore the mechanismof the effects on tumor cell growth;(3) Biochemical methods were employed to monitorproteinase activity and antioxidant levels were relative to antioxidant defense system, todetermine the relationship between the effects on intracellular metabolic processes andthat on tumor cell growth.Results:(1) Short time exposure to SMF (50-200mT) could inhibit the growth ofB16and U251, but with increasing exposure time, the effects changed ingrowth-promoting;(2) There’s no effects on cell cycle distribution after exposure toSMF (100mT,200mT);(3) The GST activity and GSH/GSSG level displayed adownward trend after the first rise, while SOD activity and total antioxidant capacitydisplayed a rise trend after first decreased, and CAT activity showed no change afterexposure to SMF (100mT,200mT) for48hours.Conclusion: Moderate-intensity static magnetic fields could inhibit the growth ofB16and U251cells, and induced intracellular oxidative stress.
