纤克颗粒对肺纤维化大鼠血清中CTGF、ILK表达影响的研究

【摘要】 目的:通过纤克颗粒对肺纤维化大鼠不同时期血清中CTGF、ILK表达影响的观察,证实纤克颗粒对肺纤维化起到防治作用。方法:本研究将建立博莱霉素诱导下大鼠肺纤维化模型。将清洁级雄性Wistar大鼠180只,随机分为6组:对照组、肺纤维化模型组、强的松组(5g/kg)、纤克颗粒高、中、低剂量组(10g/kg、5g/kg、2.5g/kg)、每组30只大鼠。使用普通专用饲料喂养。造模后第2天起开始给药,对正常对照组及肺纤维化模型组大鼠按10m1/1kg体重的标准给予生理盐水灌胃。治疗组于建立模型后第2天给予相应的药物灌胃。于连续给药后的第7天、第14天及第28天分别进行大鼠腹主动脉取血。采用ELISA测定各时期的血清中CTGF、ILK含量。结果:大鼠血清中CTGF、ILK的表达,以各时期肺纤维化模型组的表达最高,较其它组有显著的差异;强的松组与纤克颗粒高、中、低剂量组的表达低于肺纤维化模型组但高于对照组。纤克颗粒高、中剂量组能显著降低大鼠血清中CTGF、ILK的表达,以上结果提示:纤克颗粒高、中剂量组对大鼠肺纤维化有明显的防治作用。结论:纤克颗粒高、中剂量组可明显降低各时期肺纤维化大鼠血清中CTGF、ILK的含量,与强的松组相比有较明显的差异,而低剂量组与模型组相比有明显差异,但低于强的松组。实验结果说明纤克颗粒对肺纤维化有较明显的防治作用,其作用机制可能与其抑制血清中CTGF、ILK的高表达有关。

【Abstract】 Purpose: To observe the effects of XianKe Granule treat with CTGF and ILK in serumby rats which have been pulmonary fibrosis at different times, and confirm the preventiveand therapeutic effects of XianKe Granule on pulmonary fibrosis.Method: In this study, we built a model of rats with pulmonary fibrosis bybleomycin-induced. Randomly divide180Wistar male rats of clean grade into six groups,such as normal control group, pulmonary fibrosis model group, prednisone group(5g/kg),XianKe Granule high-dose group(10g/kg), middle-dose group(5g/kg) and low-dose group(2.5g/kg), each group has30rats, and treat with special feeds. Begin with the day aftermodeled the groups, administer physiological saline by1ml/100g to the rats which in normalcontrol group and pulmonary fibrosis model group, and give the relevant medicine to thetreatment groups. On7th,14th and28th day after the continuous administration, take bloodby abdominal aorta of rats, and use ELISA to measure the contents of CTGF and ILK inserum at different times.Result: he expressions show that the contents of CTGF and ILK in rats’ serum withpulmonary fibrosis model group are higher than other groups with differ significantly(p<0.01) at various periods. The expressions of Prednisone group and XianKe Granulehigh, middle and low-dose group are higher than the normal group, but lower than the modelgroup. On the7th day the expressions of CTGF in serum in order from high to low arepulmonary fibrosis model group, XianKe Granule low-dose group, middle-dose group,prednisone group, high-dose group and normal control group. On the14th day theexpressions of CTGF in serum in order from high to low are pulmonary fibrosis model group,XianKe Granule low-dose group, prednisone group, middle-dose group, high-dose group andnormal control group. The order of the expressions on the28th day from high to low also arepulmonary fibrosis model group, XianKe Granule low-dose group, prednisonegroup,middle-dose group, high-dose group and normal control group. In various periods,prednisone group compared with the XianKe Granule middle-dose group have no significantdifference, but with the other XianKe Granule groups have significant difference. Middle andlow-dose group with the high dose group are significantly different. On the7th day the expressions of ILK in serum in order from high to low are pulmonaryfibrosis model group, XianKe Granule low-dose group, middle-dose group, prednisonegroup, high-dose group and normal control group. On the14th day the expressions of CTGFin serum in order from high to low are pulmonary fibrosis model group, XianKe Granulelow-dose group, prednisone group, middle-dose group, high-dose group and normal controlgroup. The order of the expressions on the28th day from high to low also are pulmonaryfibrosis model group, XianKe Granule low-dose group, prednisone group,middle-dosegroup, high-dose group and normal control group. In various periods, prednisone groupcompared with the XianKe Granule middle-dose group have no significant difference, butwith the other XianKe Granule groups have significant difference. Middle and low-dosegroup with the high dose group are significantly different.Conclusion: XianKe Granule could decline the contents of CTGF and ILK in rats serumand has significant difference with the prednisone group, which reflect that the preventiveand therapeutic effects of XianKe Granule on pulmonary fibrosis is visible, and themechanism would relate to inhibiting the high expression of CTGF and ILK in serum.
