附子干姜配伍的药效学及毒性研究

【摘要】 目的:以有毒中药附子为切入点,选择附子配伍干姜为研究对象,考察附子干姜配伍前后对药效、毒性的影响。方法:①采用戊巴比妥钠股静脉注射法,建立大鼠急性心力衰竭动物模型,用16导生理记录仪记录血流动力学各项指标,观察配伍前后血流动力学指标的变化。②用ELISA法测定心衰大鼠血浆氨基末端脑钠肽原(NT-proBNP)、心钠素(ANP)、肾素活性(PRA)、血管紧张素Ⅱ(AngⅡ)、醛固酮(ALD)含量,观察附子干姜配伍前后对神经激素释放的调控作用。③通过测定附子、干姜单味给药与附子干姜配伍合煎液的小鼠经口LD50值,对其基础毒性进行评价和比较。结果:①以心功能为指标,对急性心衰大鼠的血流动力学考察结果表明,各给药组均可改善急性心衰大鼠血流动力学指标。附子强心作用相对缓和,与干姜配伍后,随着干姜量的增加,作用强度增强。②附子干姜配伍后均有减少大鼠血浆NT-proBNP、心钠素(ANP)、肾素活性(PRA)、血管紧张素Ⅱ(AngⅡ)、醛固酮(ALD)含量的趋势,表明附子干姜配伍后对部分神经激素的释放有调控作用。③急性毒性试验结果表明,附子LD50为17.55g/kg生药,附子:干姜1:0.5组LD50为23.46g/kg生药,附子:干姜1:1组为28.78g/kg生药,附子:干姜1:2组为32.23g/kg生药。可见,附子干姜配伍后LD50明显增大,表明毒性大大降低。结论:①附子与干姜配伍能增强治疗急性心力衰竭的作用,其作用的机理可能是通过增强心肌收缩力、改善心肌能量代谢,降低舒张末期压力,改善心脏顺应性和调控体液因子有关,从而达到回阳救逆的目的。②附子与干姜配伍可明显降低附子的毒性,提高附子用药的安全性。

【Abstract】 Purpose: Use toxic Chinese medicine aconite as the breakthrough point, choosecompatibility of aconite dried ginger as the research object, investigating the effect of efficacyand toxicity about compatibility of aconite dried ginger before and after.Methods:①Use pentobarbital sodium femoral venous injection method, establish ratacute heart failure animal model, record hemodynamic parameters by MP-150physiologicaldata recorder, observe the changes of hemodynamic parameters of compatibility before andafter.②Through ELISA kit detect the content of rat plasma NT-proBNP、ANP、PRA、AngⅡ、ALD, observe the regulation function of compatibility of aconite dried ginger beforeand after to humoral factor.③Through detect the dose LD50values in mice to evaluate andcompare the basic toxicity of compatibility of aconite dried ginger before and after.Results:①As the index to heart function, the hemodynamic investigation results ofacute heart failure rat show that all groups can improve the hemodynamic parameters of acuteheart failure in rat compatibility before and after. The cardiac contractile function relative ease,after compatibility of dried ginger, with the amount of dried ginger increased, the effectenhanced.②After compatibility of aconite dried ginger, the level of rat plasma NT-proBNP、ANP、PRA、AngⅡ、ALD down, which show that the regulation function to some humoralfactor after compatibility of aconite dried ginger.③Acute toxicity test results show that theLD50of FZ, FG1:0.5, FG1:1, FG1:2is17.55g/kg crude drug,23.46g/kg crude drug,28.78g/kg crude drug and32.23g/kg crude drug. It is clearly that after compatibility the LD50increased significantly, which indicate that the toxicity greatly reduced after compatibility.Conclusions:①After compatibility of FG, the curative effect to acute heart failureenhanced, the mechanism maybe related to enhance force of cardiac muscle systole、improvecardiac energy metabolism、lower left ventricular end-diastolic pressure、improving the heartcompliance and regulate humoral factor, and then achieve the restoring yang and rescuingpatient from collapse purpose.②Dried ginger can reduce the toxicity of aconite and improvethe safety of medication.
