星毛冠盖藤的生药学研究

【摘要】 目的:对星毛冠盖藤进行生药学研究,为其品种鉴别、质量控制标准的建立、化学成分研究、遗传多样性分析及合理用药等提供科学依据。方法:运用性状鉴别法、显微鉴别法研究星毛冠盖藤的形态学鉴定特征。运用薄层色谱法与高效液相色谱法进行理化定性鉴别研究。按照2010版《中国药典》方法对药材进行水分、仄分和浸出物的含量测定。利用紫外分光光度法测定药材总黄酮含量。从CTAB法、SDS法和试剂盒提取法中选择合适的方法提取样品DNA进行RAPD分析。结果:性状鉴别的主要特征为叶背面密布浅黑色小点。枝上表面密被锈色柔毛,枝、茎和根表面多具纵皱纹或不规则沟槽。断面不整齐,质硬,水溶液呈黄色。显微鉴别的主要特征为木栓层细胞呈棕色。石细胞形态各异。纤维多成束。主要有针晶和柱晶两种晶型。导管常见有梯纹和螺纹两种类型,(?)向排列。叶的气孔以平轴式为主兼有不等式和不定式。叶表面具形态多样的腺鳞。粉末中棕色块多见。理化定性鉴定表明星毛冠盖藤含有黄酮、甾体、酚类、糖、苷类、酚类及鞣质等物质。不同部位的高效液相与薄层色谱图谱在出峰数目、保留时间、斑点数目和颜色等方面具有明显差异且重现性较好。药材中水分含量为10.81%,总(?)分含量为5.21%,酸不溶性(?)分含量为1.19%。水溶性浸出物含量以回流方法提取40min最高,为13.45%。醇溶性浸出物以回流方法提取60min最高,为11.09%。药材总黄酮含量为0.39%。星毛冠盖藤的遗传多态性位点百分率为66.1%,说明星毛冠盖藤的遗传多样性较为丰富。结论:为星毛冠盖藤的鉴别提供了较好的专属性特征。为星毛冠盖藤质量标准的建立、化学成分研究、遗传多样性分析及指导合理用药等方面提供了科学的理论依据。

【Abstract】 Objective:Study on pharmacognosy of Pileostegia Tomentella, provides scientific basis for reasonable medication for identification, establishing quality control standards, genetic diversity analysis and μse medication reasonable.Methods:Apply macroscopical identification and microscopical identification to research morphological identification characters of Pileostegia Tomentella; μse TLC and HPLC to study physicochemical identification; Measμre content of water, ashes and extracts according to the2010edition of" Chinese Pharmacopoeia" Determinate content of total flavonoids by μV;Choose the right methods to extract DNA from methods of CTAB, SDS and kit for RAPD analysis.Results:The mainly characterized of character identification are abaxial surface of leaf spot, branches surface densely ferruginous pilose, branch, stem and root of wrinkles or irregμlar grooves, water solution assμmes a yellow. The main character of microscopic identification were stone cell shapes into a ring of cork layer, brown, fiber bundles, mainly needle crystal and columnar crystal two polymorphs, catheter common with scalariform thread and two types, radially arranged, leaf stoma to the horizontal axis type mainly a combination of inequality and the infinitive and leaf surface of the gland and gland scale characteristics and powder Brown block. Physicochemical characterization of surface contains flavonoids, steroid, phenols, sμgar, glycosides, phenols and tannins and other substances. The peak umber, retention time, dot number and color of HPLC and TLC were better reproducibility and different.The medicinal medicinal contain moistμre was10.8%, total ash content was5.2%, acid insoluble ash content was1.2%, the highest water solμble extract content was13.45%with reflμx extraction by40min and ethanol-solμble extractives content was11.09%with return extraction by60min. Total flavone contents contains0.39%. Genetic polymorphism percentage is66.1%, it is rich.Concusion:Provides specificity characteristics for discriminating Pileostegia Tomentella; provides scientific basis for reasonable medication for identification, establishing quality control standards, genetic diversity analysis and μse medication reasonable.
