小果博落回叶质量标准及其不同产地指纹图谱研究

【摘要】 小果博落回Macleaya microcarpa (Maxim.) Fedde为罂粟科博落回属植物,主要分布于陕西、甘肃等省,其果、叶、根中含有的生物碱主要为原阿片碱、别隐品碱、血根碱、白屈菜红碱四种生物碱,具有抗菌、抗肿瘤、增强机体免疫力、改善肝功能等作用。目前,同科本药材博落回提取物已在养殖生产中已作为兽药和生物农药被广泛使用,该药材中的生物碱从结构类型和总含量基本与博落回一致。为规范小果博落回叶的质量,本课题具体研究内容及结果包括以下三部分:一、小果博落回叶药材的质量标准研究本实验以小果博落回叶药材为研究对象,按照《中国药典》(2010版)一部要求,分别系统地从来源、性状、鉴别、一般杂质检查(水分、灰分、浸出物、重金属检测)、含量测定等方面对药材进行了质量研究。其中在含量测定研究方面,确定了对小果博落回叶中原阿片碱、别隐品碱、血根碱、白屈菜红碱四种生物碱的最佳提取方法,并建立了对其的HPLC检测方法,为药材质量控制标准的制定提供了参考。二、不同产地小果博落回叶药材的HPLC指纹图谱研究本实验通过对小果博落回叶药材的HPLC指纹图谱条件研究及数据分析,得出17个不同产地小果博落回叶药材的指纹图谱与对照指纹图谱的相似度均大于0.9,最终建立了小果博落回叶药材的标准指纹图谱。三、小果博落回叶与博落回叶中四种生物碱的含量比较本实验采用HPLC法对博落回叶与小果博落回叶中四种生物碱-原阿片碱、别隐品碱、血根碱、白屈菜红碱的含量进行比较研究。结果表明博落回叶中原阿片碱与别隐品碱含量较高,而小果博落回叶中血根碱与白屈菜红碱含量较高。最终本课题制定了小果博落回叶的质量标准,得到了其不同产地的对照指纹图谱,并区别了易混淆品博落回叶,为更准确、更全面地评价小果博落回叶的质量提供了依据。

【Abstract】 Macleaya microcarpa (Maxim) Fedde is a species of the genus Macleaya R. Br., Papaveraceae sp..It is widely distributed in Shanxi、Gansu、Hubei in China.The main active ingredients are protopine, allocryptopine, sanguinarine, chelerythrine.It has important value in anti-bacterial, anti-tumour, improve liver function and Enhance immunity and so on.At present,the herb extract is largely used in breeding production as a Veterinary Drugs and biological pesticide.For ensuring the leaf quality of Macleaya microcarpa(Maxim)Fedde,the main contents and results were divided into three sections as follows:(1) The quality standard research of the leaf of Macleaya microcarpa (Maxim) FeddeThe topic of the experiment studies is leaf of Macleaya microcarpa (Maxim) Fedde.According to technical requirements in the appendix in China Pharmacopoeia (I,2010ED),systematically to study the topic through source, characters, identification, inspection (moisture, ash content, extract, heavy metal detection),the content determination and so on.The research set up a best extraction method and High performance liquid chromatography (HPLC) analysis method for protopine, allocryptopine, sanguinarine, chelerythrine in the content determination, and provide a reference of qualifying control methods for medicinal formulation to provide a reference. (2) The HPLC fingerprint research of the different origin of Macleaya microcarpa (Maxim) Fedde.The experiment study the HPLC fingerprint conditions and data;establish the standard fingerprint and obtain the similitude degree between the leaf of Macleaya microcarpa (Maxim) Fedde in17different origins and the standard fingerprint more than0.9.(3) Comparison of contents of four alkaloids in leaf of Macleaya microcarpa (Maxim) Fedde and Macleaya Cordate (Willd.) R. Br. HPLC method was used for the determination of four alkaloids in leaf of Macleaya microcarpa (Maxim) Fedde and Macleaya Cordate (Willd.) R. Br..We found that the content of protopine and allocryptopine in leaf of Macleaya Cordate (Willd.) R. Br. is more than in Macleaya microcarpa (Maxim) Fedde, but content of sanguinarine and chelerythrine.Finally, we establish the leaf quality standard and standard fingerprint of Macleaya microcarpa (Maxim) Fedde, distinguish Macleaya Cordate (Willd.) R. Br. that is easily confused, and provides the basis for evaluating the leaf quality of Macleaya microcarpa (Maxim) Fedde more accurately and comprehensively.
