铁皮石斛多糖改善D-半乳糖诱导小鼠衰老作用研究

【摘要】 目的:探讨铁皮石斛多糖(PDC)对D-半乳糖诱导小鼠衰老作用的影响及其可能机制。方法:选取健康昆明种小鼠90只,随机分为正常对照组、衰老模型组、健延龄400mg/kg组、PDC40mg/kg、80mg/kg和160mg/kg组,每组15只。各给药组小鼠除每天颈背部皮下注射D-半乳糖120mg/kg外,同时灌胃给予相应的药物,连续5周用药后检测各组小鼠的学习记忆能力,用药6周后检测各组小鼠的胸腺和脾脏重量指数、血清、肝和脑组织中丙二醛(MDA)、超氧化物歧化酶(SOD)及谷胱甘肽过氧化物酶(GSH-PX)含量。结果:经PDC处理的小鼠,虽可增加穿越原平台的次数,但仅见在第四象限游泳时间和路程可明显延长(p<0.05或p<0.01),也可延长小鼠第一次跳下平台的潜伏期和减少触电次数(p<0.05或p<0.01)。同时可增加胸腺和脾脏重量指数,增加血清、肝和脑组织中SOD和GSH-PX活性,降低MDA含量(p<0.05或p<0.01)。结论:PDC能够改善D-半乳糖诱导衰老小鼠的学习记忆能力,提示其有一定的抗衰老作用,可能的机制与通过增加机体SOD和GSH-PX活性,促进氧自由基的清除,以及增强机体的免疫功能有关。

【Abstract】 Aim: To observe the effects of polysaccharides from Dondrobium candidum (PDC)on mouse aging induced by D-galactose and investigate its possible mechanisms.Methods:90kunming mice were randomly divided into6groups (n=15), namely,control group, model group, JYL400mg/kg group, PDC40,80and160mg/kg groups.An aged mouse model was induced by subcutaneous injection of D-galactose120mg/kg when these mice were simultaneously treated with medicine by gavage. Theabilitiy of mouse learning and memory was examined after treatment with medicine for5weeks, the weight indexes of thymus and spleen, the activities of superoxidedismutase (SOD) and glutathione peroxidase (GSH-PX) as well as level ofmalondialdehyde (MDA) in serum, brain and liver were measured after treatment withmedicine for6weeks.Results: After treatment with PDC, the number of across original platform had anincrease trend, the time and distance of the fourth quadrant swim were significantlyincreased(p<0.05or p<0.01), the latent period that mice first jumped down platformwas significantly delayed, and the number of electric shock was significantly decreasedas well(p<0.05or p<0.01). On the other hand, the weight indexes of thymus andspleen, the SOD and GSH-PX activities in serum, brain and liver were significantlyincreased, while the MDA content in serum, brain and liver was significantly decreased(p<0.05or p<0.01).Conclusion: PDC could improve the ability of learning and memory in aged miceinduced by D-galactose, which suggested that PDC possessed the antiaging effect, andits mechanisms might be related to enhancing the SOD and GSH-PX activities,subsequently scavenging the oxygen free radicals, and increaseing the immune function.
