当归药渣有效成分的提取纯化及综合利用研究

【摘要】 本文对当归药渣中多糖及阿魏酸的提取、纯化工艺及药理作用进行研究和优化,为其二次开发利用提供技术方案和实验数据。1.通过对当归药渣中多糖的提取纯化工艺的探索,确定水提和醇沉的最佳工艺路线。采用单因素试验和正交实验优化多糖提取工艺,获得最佳提取工艺为按料液比1:8在100℃下提取3次,每次2h;最佳醇沉工艺为按浓缩比1:1,用95 %乙醇沉淀多糖,使醇浓度最终为80 %,低温放置6h后,离心,得粗多糖沉淀物。2.在多糖纯化过程中,对脱蛋白次数及脱色进行研究,选取Sevag法脱蛋白共3次,0.1 %活性炭脱色即可。采用分级沉淀法得到当归精制多糖Ⅰ(ASPⅠ)和当归精制多糖Ⅱ(ASPⅡ)。采用苯酚-硫酸法测定3种多糖的含量和得率,当归粗多糖含量50.84 %,得率10.73 %;当归精制多糖Ⅰ含量62.84 %,得率7.84 %;当归精制多糖Ⅱ含量69.16 %,得率3.33 %。采用考马斯亮蓝法测定蛋白质含量,三种多糖中蛋白质含量分别为19.49 %,17.11 %和15.86 %。3.通过正交设计实验考察了乙醇浓度,提取温度,液料比和提取时间对当归药渣中阿魏酸提取效果的影响,确定醇提最佳工艺为按料液比1:16,在85℃下用80 %乙醇回流提取3次,每次2.5 h。以上条件下得到的阿魏酸的含量为0.04247 mg/g。4.当归药渣中提取的3种多糖对二甲苯引起小鼠耳肿胀及吲哚美辛引起的血管通透性有明显的抑制作用,表明当归多糖具有一定的抗炎效果,其中当归精制多糖Ⅰ的量效关系更为显著。5.选取当归精制多糖Ⅰ进行免疫功能实验,试验结果表明它对环磷酰胺所致的小鼠吞噬细胞活性,脾指数及胸指数均表现为增强作用,并能增加小鼠血清溶血素值。说明当归精制多糖Ⅰ具有一定的免疫作用。

【Abstract】 In this article, the extraction and purification craft of polysaccharide and ferulic acid from the dregs of Angelica sinensis(Oliv.)Diels were researched and optimized, and the pharmacological action of polysaccharide was researched . The objective of research is to provide the technical program and empirical datum for redevelopment use of Angelica sinensis.1. Through exploring the polysaccharide extraction and purification craft from the dregs of Angelica sinensis, it was determined the best route of water-extraction and alcohol-precipitation by the single factor experiment and the orthogonal test. Optimum parameters for the extraction with water is as follow: temperature100℃,time2h , solid-liquid ratio 1:8 and frequency ratio 3. then the extracting solution was condensed to the cocnentration factor 1:1and injected 95% alcohol to make the alcohol concentration to 80%. Thick polysaccharide was settled by offcenter under the low temperature for 6h.2. In the polysaccharide purification process, the protein-free number of times and the decolorization was researched. It was determined 3 times for protein-free number by the Sevag method and 0.1 % activated carbon for decolorization. It was obtained two kinds of purification polysaccharides from the thick polysaccharide by fractional precipitation, and determined the contents and rates of the three kinds of polysaccharides by the Phenol-sulfuric acid method, the thick polysaccharide content is 50.84 % and rate is 10.73%; the content of angelica purification polysaccharideⅠis 62.84%, rate is 7.84%; the content of angelica purification polysaccharideⅡis 69.16%, rate is 3.33%. The protein content of three kinds of polysaccharides is respectively 19.49 %, 17.11 % and 15.86% by Bradford method.3. Uniform design was adopted to observe the effect of alcohol density, temperature, time and solid-liquid ratio on ferulic acid extraction rate, and optimum parameters for the extraction with alcohol is as follow: temperature85℃, time2.5h, solid-liquid ratio1:16 and frequency ratio 3. The content of ferulic acid is 0.04247 mg/g under this condition.4. Three kinds of polysaccharides from the dregs of Angelica sinensis can control ear edema in mice which was casued by dimethylbenzen and the blood vessel permeability by the acetic acid-induced, which is indicated that the angelica polysaccharides have certain antiinflammatory effect, especially the quantity effect relations of the angelica purification polysaccharideⅠis more remarkable.5. Angelica purification polysaccharideⅠwas carried on the immunization experiment, the test result indicated that it can increase ctx-induced Phagocytosis index, Thymus index and Spleen index, and can increase the mouse blood serum hemolysin value. so the angelica purification polysaccharideⅠhas certain effect in immunization.
