【摘要】 目的观察麝香乌龙丸对兔膝骨关节炎滑膜中SOD、iNOS的表达的影响,以探讨麝香乌龙丸治疗骨关节炎可能的作用机理,为其临床应用提供试验依据。方法由小到大编号,按随机数字表随机分成四组,每组10只:A组:正常对照组;B组:模型组;C组:麝香乌龙丸组;D组:抗骨增生片组。B,C,D三组于右膝内注射木瓜蛋白酶(1.6%)生理盐水溶液建立膝骨关节炎模型。2.给药:模型成功后,C组按成人的每日每公斤体重用药量的6倍剂量,将麝香乌龙丸溶于10ml生理盐水中灌胃给药;D组按同法将抗骨增生片灌胃给药,每日一次。A、B组均以等量生理盐水灌胃,共4周。3.免疫组化法观测各组兔膝滑膜中SOD、iNOS的表达情况。结果SOD、iNOS阳性表达:SOD、iNOS的阳性表达定位于细胞浆,呈棕黄色或黄色颗粒,主要分布在滑膜衬里层和滑膜下层。B组滑膜中SOD阳性表达率较A组降低(P<0.01);C、D组滑膜中SOD阳性表达率较B组明显增高(P<0.01);C组与D组比较无统计学差异(P>0.05)。iNOS在A组无表达;C、D组滑膜中iNOS阳性表达率较B组均显著降低(P<0.01),C、D组比较无统计学差异(P>0.05)。结论本实验证明:麝香乌龙丸能上调滑膜中SOD、下调iNOS的表达。更多还原

【Abstract】 Objectives The purpose of the experiment is to observe the influence of Musk-Aconite-Earthworm Pill on SOD and iNOS in synovial membrane in rabbit knee osteoarthritis, and discuss the possible mechanism of action for curative effect of Musk-Aconite-Earthworm Pill on osteoarthritis, and therefore supply experimental basis to its clinical application.Methods 1.Divide the rabbits into 4 groups and establish model: number the 40 healthy male Japanese white rabbits according to their weight and divide them at random into 4 equal groups: Group A: normal comparison group; Group B: model group; Group C: Musk-Aconite-Earthworm Pill group; Group D: osteoproliferation resistant tablet group. And then establish model of experimental knee osteoarthritis with intra-articular injection of papain(1.6%) physiological saline solution .2.Provide medicine: provide in gastric perfusion way 6 times Musk-Aconite-Earthworm Pill solved in 10ml physiological saline solution of the quantity per day and per kg weight of for an adult to Group C; and provide osteoproliferation resistant tablet in the above method to Group D once per day. And provide Group A and Group B with equal quantity of physiological saline solution in gastric perfusion way. Continue this for 4 weeks.3.Observe the expression of SOD and iNOS by immunohistochemistry.Results Expression of SODand iNOS: The positive SOD、iNOS located in cytoplasm, brownish or brownish granule. Mainly distributed in synovial lining and synovial below. Compared with group A,expression rates of SOD in group B increased (P<0.01),compared with group B, SOD in group C and D increased(P<0.01),The study showed no statistically significant in group C with group D (P>0.05);iNOS is no expression in group A,Compared with group B, iNOS in group C and D decreased (P<0.01), The study showed no statistically significant in group C with group D (P>0.05).Conclusion Musk-Aconite-Earthworm pill can up-regulate the expression of SOD and down-regulate the expression of iNOS.更多还原
