【摘要】 目的：探讨清肺口服液对人喉癌上皮细胞(Hep-2)感染呼吸道合胞病毒(RSV)后细胞因子白细胞介素-8(IL-8)、细胞间粘附分子-1(ICAM-1)表达的影响。方法：分时段以清肺口服液含药血清,利巴韦林含药血清及空白血清干预RSV感染的体外培养的Hep-2细胞,48小时及72小时后收集各组细胞上清,采用酶联免疫吸附试验法(ELISA法)测定各组Hep-2细胞IL-8、ICAM-1表达的变化。结果：清肺口服液含药血清对Hep-2细胞的最大无毒浓度为20%。48小时及72小时后病毒模型组IL-8、ICAM-1的表达明显高于清肺口服液组(Ⅰ、Ⅱ、Ⅲ)、利巴韦林组(Ⅱ、Ⅲ),有显著性差异；空白血清组(Ⅱ、Ⅲ)与病毒模型组无显著性差异。清肺口服液组(Ⅰ、Ⅱ、Ⅲ)与利巴韦林组(Ⅱ、Ⅲ)比较,无显著性差异。清肺口服液Ⅰ、Ⅱ、Ⅲ组之间比较,无显著性差异。72小时后各组IL-8、ICAM-1的表达与48小时相比,无显著性差异。结论：清肺口服液可降低人喉癌上皮细胞(Hep-2)感染RSV后细胞因子IL-8、ICAM-1的过度表达,减轻炎症反应,提前给药更可获得良好的效果,这可能是清肺口服液抗病毒的作用机制之一。总言之,清肺口服液可以拮抗RSV病毒,清肺口服液是治疗RSV感染的有效复方制剂。更多还原

【Abstract】 Objective:To investigate the effects of Qingfei oral liquid on expression of Interleukin (IL-8 and Intercellular adhesion molecules (ICAM)-1 in Human epithelial cells (Hep-2) infected with Respiratory Syncytial Virus (RSV)Methods:Hep-2 cells were intervened with serum containing Qingfei oral liquid, Ribavirin and blank serum, respectively. In each of these three groups, intervention was conducted before, simultaneously or after RSV infection (Ⅰ,Ⅱ,Ⅲ). Expression levels of IL-8 and ICAM-1 were detected by enzyme linked immunosorbent assay (ELISA), after collecting the cellular supernatant.Results:Qingfei oral liquid containing serum on Hep-2cells maximum non-toxic concentration of 20%. After 48 and 72 hours of cultivation, expression levels of IL-8 and ICAM-1 in virus control group were significantly higher than those in Qingfei oral liquid groups (Ⅰ,Ⅱ,Ⅲ) or Ribavirin group (Ⅱ,Ⅲ). No significant difference was found between blank serum control groups (Ⅱ、Ⅲ) and the virus control group. Difference between the Qingfei oral liquid groups (Ⅰ,Ⅱ,Ⅲ) and the Ribavirin groups (Ⅱ,Ⅲ) was insignificant either. Expression levels of IL-8 and ICAM-1 within the Qingfei oral liquid group (Ⅰ,Ⅱ,Ⅲ) were slightly different after 48 or 72 hours of cultivation.Conclusion:Qingfei oral liquid can decrease the over-expression of IL-8 and ICAM-1 so as to relieve inflammation. Furthermore, better effects can be achieved when it was administrated in early stage of RSV infection. These foundings may relate to the antiviral mechanismRemember of Qingfei oral liquid. Qingfei oral liquid can antagonize RSV, and it is an effective and safe compound in treating RSV infections.更多还原
