针刺督脉经穴对脑缺血再灌注大鼠缺血半暗带Caspase-3、TNF-α和IL-1β蛋白表达的影响

【摘要】 目的:观察针刺督脉经穴对脑缺血再灌注大鼠缺血半暗带Caspase-3、TNF-α及IL-1β蛋白表达的影响;探讨针刺督脉经穴对缺血再灌注损伤脑组织的保护作用机制。方法:以局灶性脑缺血再灌注大鼠模型为缺血性脑血管病研究对象,以针刺督脉经穴为干预手段,并与传统针刺法(针刺阳明经穴组)进行对照,观察干预治疗前后模型大鼠神经功能评分的变化,同时采用Western blot方法检测各组大鼠在干预治疗前后脑组织中关键凋亡执行蛋白酶Caspase-3及炎症细胞因子TNF-α和IL-1β蛋白表达的变化情况。结果:(1)神经功能评分:针刺督脉穴组与针刺阳明经穴组治疗前后评分均有显著性改善(P<0.05),针刺督脉穴组较针刺阳明经穴组可更显著的改善大鼠缺损的神经功能(P<0.05)。(2)Caspase-3、TNF-α和IL-1β的蛋白表达:假手术组大鼠Caspase-3、TNF-α和IL-1p在脑组织呈基础水平表达;模型组Caspase-3、TNF-α和IL-1β较正常组显著增加(P<0.01),呈高水平表达;针刺督脉经穴和针刺阳明经穴组与模型组比较均显著降低(P<0.01或P<0.05),但仍高于假手术组和正常组,但针刺督脉经穴组较针刺阳明经穴组有更显著的效果(P<0.05)。结论:针刺督脉经穴能够更显著的改善脑缺血再灌注后大鼠的神经功能损伤;且较针刺阳明经穴组可更显著的降低脑缺血再灌注大鼠脑组织Caspase-3、TNF-α和IL-1β蛋白的表达,从而达到拮抗缺血性脑损伤的作用,说明针刺督脉经穴拮抗脑缺血再灌注损伤脑组织的神经元凋亡机制可能与其更显著的抑制脑内Caspase-3的蛋白表达有关,而抑制炎症反应的机制可能与其抑制TNF-α和IL-1β蛋白的过度表达有关。

【Abstract】 Objective:To observe the effect on protein expression of Caspase-3,TNF-a and IL-1βin ischemic penumbra of cerebral ischemia reperfusion rats treated by acupuncturing DU meridian;and to explore the mechanisms of acupuncturing DU meridian in protecting ischemia reperfusion tissue.Methods:Focal cerebral ischemia rats model were used for the object of research,and they were treated separately by acupuncturing DU meridian acupoints and Yangming meridian acupoints.Then the changes of neurobehavioral scores were observed.Besides,the changes of the expression of protein Caspase-3,TNF-a and IL-1βwere detected by Western blot method before and after the treatment.Results:(1)neurological score:Both the DU meridian acupoints acupuncture and the Yangming meridian acupuncture can significantly improve the neurological score before and after treatment compared with the model group(P<0.05), and the DU meridian acupoints acupuncture group has the better effect than the Yangming meridian acupuncture group(P<0.05). (2)Protein expression of Caspase-3、TNF-αand IL-1β:Caspase-3、TNF-αand IL-1βin the brain tissue of the sham operation rats are on the level expression;and they are all significantly higher expressed in the model group than in the normal group(P<0.01).In the DU meridian acupoints acupuncture and the Yangming meridian acupuncture group,all the three protein expressions are lower than the model group(P<0.01&P<0.05),but higher than the sham operation and the normal,and the DU meridian acupoints acupuncture group has a more significant effect.(P<0.05).Conclusion:Acupuncturing DU meridian acupoints can more significantly improve the neurological damage after ischemia-reperfusion injury of rats,and has more significantly effect on reducing the protein expression of Caspase-3,TNF-a and IL-1βthan acupuncturing the Yangming meridian to achieve the purpose of anti-ischemic brain injury.This maybe the mechanisms of acupuncture protecting the injury brain.
