丹槐银屑浓缩丸对豚鼠银屑病样模型IL-8和IL-10影响的研究

【摘要】 目的:研究丹槐银屑浓缩丸对豚鼠银屑病模型IL-8和IL-10的影响,探讨其治疗银屑病的相关作用机制。为丹槐银屑浓缩丸治疗银屑病提供可靠的科学依据,同时为其在临床的广泛应用提供理论基础。方法:5%普萘洛尔外涂豚鼠耳部皮肤,使其产生银屑病样病理改变,然后分别以丹槐银屑浓缩丸、复方青黛胶囊和甲氨喋呤注射液治疗,光镜下观察局部皮肤病理变化,ELISA法检测皮损组织IL-8和IL-10水平,观察该变化与皮损表现的关系。结果:普萘洛尔致豚鼠银屑病模型血清和组织中的IL-8水平高于正常组(P<0.01),IL-10水平明显低于正常组(P<0.01)。MTX组、青黛组及丹槐高、中、低三个剂量组均能有效降低IL-8水平;提高IL-10水平,治疗效果明显,与模型组比较差异有显著性(P<0.05,P<0.01)且MTX、青黛胶囊和丹槐中剂量的疗效最好。结论:丹槐银屑浓缩丸对普萘洛尔致豚鼠耳背银屑病样模型有治疗作用,其作用机理可能与其影响IL-8、IL-10水平,改变Thl/Th2的平衡有关。

【Abstract】 Objective:To explore the effects of DAN HUAI YIN XIE Concentrated Pill on the Expression Levels of IL-8 and IL-10 in Psoriasis-like Lesions of Guinea Pigs Caused by Propranolol. Explore the mechanism related to the treatment of psoriasis. Provide reliable scientific basis,and for it’s extensive application provide theoretical basis.Methods:5% propranolo emulsion was applied on ears back of Guinea pigs to produce the psoriais-like models,then respectively treat with experimental drug. Light microscope to observe the pathological changes of local skin. Tissue lesions detected IL-8 and IL-10 levels by ELISA. Observe the performance levels and the relationship between skin lesions.Results:Guinea pig model of psoriasis induced by propranolol in serum and tissue levels of IL-8 higher than the normal group (P<0.01), IL-10 levels were significantly lower than normal group (P<0.01). MTX group, QINGDAI group and DANHUAN high, medium and low dose groups could effectively reduce the level of IL-8; increased levels of IL-10, significant treatment effect. Compared with the model group were significantly different (P<0.05, P<0.01). Furthermore, the efficacy of the best is MTX, QINGDAI capsule and DANHUAI medium dose.Conclusions:DAN HUAI YIN XIE Concentrated Pill have a therapeutic effect on psoriasis-like lesions of guinea pigs’ears caused by propranolol.and the mechanism may be related to influence IL-8, IL-10 levels, changing the balance of Thl/Th2.
