鼻窍通汤药学及药效学实验研究

【摘要】 目的:验证鼻窍通汤与功能主治有关的药效学作用材料与方法:1材料:1.1鼻窍通汤剂辽宁省中医药研究院制剂中心提供,批号:100802。1.2 SPF级昆明种小鼠,是由中国医科大学实验动物部提供的,动物许可证号:SCXK（辽）2008-0005。1.3SPF级Wistar的大鼠是由中国人民解放军军事医学科学院的实验动物中心提供,动物许可证号:SCXK-（军）2007-004。1.4藿胆丸,由广州王老吉药业股份有限公司生产,生产批号:1008151.5消旋山莨菪碱片,由杭州民生药业集团有限公司生产,生产批号:T08H290;1.6阿司匹林片,由维康医药集团沈阳延风制药有限公司生产,生产批号:20070901;1.7鼠颗粒饲料,由北京科澳饲料有限公司生产,许可证号:SCXK（京）2009-0012。2方法:2.1由于无法测出鼻窍通汤小鼠灌胃给药的半数致死量(LD₅₀),故以鼻窍通汤所能配成的最大浓度和小鼠所能承受的最大灌胃给药容积,进行小鼠灌胃给药的最大给药量的测定。2.2化学药物反复刺激引起动物慢性鼻炎法考察药物的治疗作用。2.2.1小鼠鼻炎模型的建立。2.2.2对鼻炎模型小鼠炎症组织中组胺含量的影响。2.2.3利用肾上腺素造成小鼠肠系膜微循环障碍,观察药物对小鼠肠系膜微循环的影响;2.3.4大鼠皮下注射盐酸肾上腺素注射液0.8ml/kg,分两次注射,期间用冰水刺激,造成大鼠血瘀模型,观察鼻窍通汤对大鼠血液流变学的影响;2.3.5考察药物对不同菌株的敏感性,即采用用测定抑菌圈直径的方法,即抗菌作用;结果:1.鼻窍通汤在所能配成的最大浓度1.14g/ml和小鼠所能承受的最大灌胃给药容积40ml/kg的情况下,一日内给小鼠灌胃给药的最大给药量为45.6g（生药量）/kg,为成人每日口服量的142倍。（成人每日口服量为19.2g（生药量）,成人体重按60kg计）;2.鼻窍通汤能够明显降低鼻炎模型小鼠炎症组织中组胺含量;3.鼻窍通汤能明显改善小鼠肠系膜微循环障碍;4.鼻窍通汤能显著降低血瘀模型大鼠全血比粘度、血浆比粘度、红细胞压积及红细胞电泳时间。5.鼻窍通汤能对所选菌株的抑菌圈直径均大于15mm。结论:1.鼻窍通汤能降低小鼠鼻炎模型炎症组织中的组胺含量,对慢性鼻炎有明显的对抗作用;2.鼻窍通汤中、高两个剂量组均明显提高滴加肾上腺素后8min的流速,改善流态,同时增加血管直径,与空白对照组比较均有显著性差异（P﹤0.05,P﹤0.01）,提示鼻窍通汤具有改善小鼠肠系膜微循环的作用,即鼻窍通汤有改善微循环的作用。3.鼻窍通汤能使大鼠降低血浆粘度及红细胞压积,减少纤维蛋白原含量,缩短红细胞电泳时间,与模型组比较差异显著（P﹤0.05,P﹤0.01）。表明鼻窍通汤可使血瘀模型之粘、浓、凝、聚明显恢复,提示鼻窍通汤有活血作用。4.鼻窍通汤能使肺炎链球菌、铜绿假单胞菌抑菌圈直径大于19 mm,金黄色葡萄球菌、大肠埃希氏菌、无乳链球菌、化脓链球菌、痢疾志贺菌对鼻窍通汤中度敏感,抑菌圈直径介于13～19mm之间,提示鼻窍通汤具有抑菌作用。更多还原
