分子印迹聚合物制备方法研究及性能评价

【摘要】 分子印迹技术是对目标分子具有特定识别能力的一种聚合物制备技术,由于具有与天然抗体相似的识别选择性和高分子材料的抗腐蚀性的双重优点,在色谱固定相、固相萃取、仿生化学传感器、手性拆分等方面展现了广泛的应用前景。本文对分子印迹聚合物的制备方法及性能进行研究,具体内容如下:采用本体聚合和原位聚合法制备乙胺嘧啶印迹聚合物。通过紫外光谱法对功能单体及用量进行选择,用静态吸附及高效液相色谱的方法对聚合物进行评价,结果表明印迹聚合物对模板分子具有较高的特异选择性,实现了乙胺嘧啶与其结构类似物敌菌净基线分离。表面印迹技术制备N-Boc-L-色氨酸印迹聚合物。通过静态平衡结合法以及Scatchard分析法研究了该聚合物的结合性质和选择性能。与空白聚合物相比,印迹聚合物具有较高的吸附效率和选择性,印迹效率Eia为70.67%,远高于本体聚合方法的印迹效率,将所得的聚合物用作高效液相色谱固定相,实现了外消旋混合物N-Boc-DL-色氨酸的基线分离。以牛血红蛋白为模板分子,采用溶胶-凝胶法制备分子印迹聚合物。对三种洗脱液[10%(V/V)HAC-10%(m/V)SDS,2mol/LNaOH,30%(V/V)HAC]进行选择优化,实验结果表明2mol/LNaOH洗脱效果较好,通过对比洗脱率与吸附量的关系曲线表明,当洗脱率在40%-60%时,吸附量达到最大值。利用静态吸附法研究了印迹聚合物的吸附特性,为蛋白质的立体印迹提供理论与实验参考。

【Abstract】 Molecular imprinting technology is a polymer preparation technology fou target molecules have specific recognition ability, In the chromatogram immobility, solid phase extraction, the bionic chemical sensors, chiral resolution aspects show the broad application prospect because of having double merit which are natural antibodies similar identifying selectivity and polymer materials corrosion resistance.In this paper, the preparation methods of molecularly imprinted polymer and performance were studied, Specific content as follows:Pyrimethamine molecularly imprinted polymer was synthesized with bulk polymerization and in-situ polymerization. functional monomers and dosage were chose by ultraviolet spectrometry, polymer were evaluated by static adsorption and HPLC, Results show that molecularly imprinted polymer has high specific selectivity for template molecule, Realizing the baseline separation for pyrimethamine and diaveridine which is structural analogues of pyrimethamine.N-Boc-L-tryptophan was synthesized by Surface imprinting technique.Its combination and select properties were researched by static balance and Scatchard analysis method . Molecularly imprinted polymer with higher adsorption efficiency and selectivity compare with blank polymer, Imprinting efficiency Eia was 70.67% which is much higher than bulk polymerization methods, The polymers is used as HPLC stationary phase, Realizing the baseline separation for racemic mixture N-Boc-DL-tryptophan.Use bovine hemoglobin as template molecules, Molecularly imprinted polymer was synthesized by sol-gel. Selection optimization [10%(V/V)HAC-10%(m/V)SDS, 2mol/LNaOH, 30%(V/V)HAC], Experimental results show that elution efficiency of 2mol/LNaOH is the best, Through comparing the elution rate with the adsorption quantity of relationship curves indicate that the adsorption quantity reach maximum when elution rate was 40%-60%. Adsorption characteristic of molecularly imprinted polymer was research by static adsorption method which provide theoretical and experimental reference for protein three-dimensional imprinting.
