补肾填精方对再障模型小鼠骨髓造血干/祖细胞CFU-E、CFU-GM影响的实验研究

【摘要】 目的:观察补肾填精方药对再障模型小鼠骨髓造血干/祖细胞增殖、骨髓病理及外周血细胞的影响,探讨补肾填精方药治疗再障的作用机理。方法:昆明种雄性小鼠腹腔注射环磷酰胺及皮下注射苯油造模后,经灌胃给予补肾填精方悬液35天后眶动/静脉采血法检测外周红细胞(RBC)、血红蛋白(Hb)、白细胞(WBC)、血小板(PLT)的量,并处死小鼠取股骨骨髓制备有核细胞悬液进行有核细胞(BMNC)计数,甲基纤维素半固体法培养红系祖细胞集落(CFU-E)及粒-巨噬细胞集落(CFU-GM)并计数,观察补肾填精方对再障模型小鼠造血祖细胞影响;取小鼠胸骨用石蜡包埋法镜下观察骨髓造血组织情况。结果:补肾填精方中、高剂量组小鼠RBC、Hb、WBC、PLT较模型组升高,有统计学意义(P<0.05);补肾填精方低、中、高剂量组BMNC、CFU-E及CFU-GM数目较模型组增多,高剂量组与中剂量组间上述指标存在差异,且有统计学意义(P<0.05),高剂量组与中剂量组间上述指标无差异(P>0.05);骨髓病理显示:模型组小鼠骨髓腔充满脂肪组织,造血结构破坏,补肾填精方组脂肪细胞明显减少,三系细胞增生,且以高剂量组较为显著。结论:补肾填精方可使骨髓造血干/祖细胞CFU-GM、CFU-E增殖,修复造血组织结构,提升外周血细胞作用,通过对造血干/祖细胞的影响而发挥作用。

【Abstract】 Objective:To investigate the effects of Bushentianjingfang on Bone marrow hemopoietic progenitor cells and Bone marrow pathology and Peripheral blood cells in mice with Aplastic Anemia. Methods:Mading mouse model of aplastic anemia by injecting kunming mice on Abdominal with Cyclophosphamide and on Subcutaneous with benzene-cornoil,affert mading successful medle mice,giving the above mice bushentianjingfang suspension for 35days, the blood was taken from theorbital arteriovenous and peripheral blood cells were counted; The mices were sacrificed under the sterile condition, left femur was taken and Bone marrow mononuclear cells was prepared, the number of bone marrow mononuclear cells(BMNC) were counted. CFU-E and CFU-GM were measured by methylcellulose seni-solid colony culture,to observed the effect of bushentianjingfang on hematopoietic progenitor cells in mice. Results:RBC Hb WBC PLT in bushengtianjingfang high and middle dosage treated group were higher than that in the AA group (P<0.05); BMNC CFU-E CFU-GM in bushengtianjingfang treated group were higher than that in the AA group, there is differences between bushengtianjingfang high and low dosage treated groups (P<0.05),but on in high and middle group(P> 0.05);Model group mice marrow cavity was filled with fat tissue and hematopoietic structure was damaged. The number of fat cells were decreased and three series cells were increased,the high dosage group was significant.Conclusion:Bushentia-njigfang can improved the numbers of hemopoietic progenitor cells and peripheral blood cells.at the same time, hematopoietic tissue was repaired;Bushentianjingfang played a role with the influence of hemopoietic progenitor cells.
