长川霉素产生菌的选育和生防菌株AⅠ的特性研究及鉴定

【摘要】 长川霉素是一种大环内酯类结构的农用抗生素杀菌剂,也是一种微生物来源的免疫抑制剂,被广泛的应用于农药和免疫学领域,具有很好的经济应用价值。本文采用了紫外诱变、N+注入诱变、B+注入诱变、紫外线-氯化锂复合诱变和紫外线-亚硝酸复合诱变等5种诱变方法,对长川霉素产生菌SPRI-2098先后进行了四轮诱变,筛选到生长周期较短,目标产物长川霉素产量高,且发酵条件要求不高的突变株,编号为Ⅳ-90,其效价为292.129ppm,比原始菌株的效价114.249ppm提高了155.70%。另外,还对一株在盆栽筛选实验中筛选得到的生防菌株AⅠ进行了部分生理生化特性研究和分类鉴定。该菌株对灰霉病菌有100%防治效果、对白粉病菌有70.9%防治效果。经过生理生化试验和16S rDNA核苷酸序列的测定,鉴定结果显示,该菌株属于枯草芽孢杆菌属第Ⅰ类的一种枯草芽孢杆菌。

【Abstract】 As one kind of agricultural fungicide with a macrolide antibiotic structure and also a immune inhibitors from Microbiology, Ascomycin has good economic value which is widely used in pesticides and immunology.In this study, five mutation methods was carried out on the Ascomycin producing strain SPRI-2098, which include UV mutation, N+implantation, B+implantation, UV-LiCl mutation and UV-nitric acid mutation. Through four rounds of mutation experiments and screening, one mutant NO.Ⅳ-90 with high production of Ascomycin was yield, whose titer is 292.129ppm, increased by 155.70% than the original strain titer 114.249ppm.Furthermore, one biocontrol bacteria A I selected from potted plant experiment was identified, which has 100% control efficiency to gray mold and 70.9% to powdery mildew respectively. It is finally identified as one species of Bacillus subtilis based on physiological biochemical character and 16SrDNA sequence analysis.
