降解HNS生产废水的复合型微生物絮凝剂产生菌筛选及应用研究

【摘要】 本论文从六硝基茋生产废水、被六硝基茋生产废水污染的土壤和杨树枝上,经过分离筛选驯化,得到几种微生物絮凝剂产生菌,经复合最后筛选出一种较高效的复合型微生物絮凝剂产生菌;通过单因素实验确定其最佳的培养条件和絮凝条件,将其应用到六硝基茋生产废水的处理中,并进行絮凝活性分布的测定,对所产复合型微生物絮凝剂进行了分离纯化及初步鉴定,实验结果表明:共得到15种纯种菌落从六硝基茋生产废水及被其污染的土壤、杨树枝中用六种富集培养基经多次分离、纯化得到,经驯化和对其絮凝率的测定,获得7株耐六硝基茋废水的具有絮凝活性的高效菌种,经驯化它们的絮凝率均发生了变化,有增有减,这说明驯化对微生物絮凝剂产生菌的产絮能力有一定的影响,选择5株具有较高絮凝活性的菌种进行复合,得到絮凝率大大高于单一菌群的复合菌群MBF4,其絮凝率达77.60%。复合型微生物絮凝剂产生菌MBF4的培养基经优化其最佳碳源为复杂糖淀粉,最佳氮源为蛋白胨,絮凝剂产生的最适宜pH值为5。复合型微生物絮凝剂产生菌MBF4对高岭土悬浊液的絮凝条件经优化,其最佳絮凝条件为:投加量3mL,pH值8,金属离子为Ca2+。复合型微生物絮凝剂产生菌MBF4在处理六硝基菧生产废水时最佳投加量为6ml,最佳pH值为8,对六硝基茋生产废水色度、COD的去除率可达93.33%、73.08%,为六硝基茋生产废水的后续处理提供了有利条件。复合型微生物絮凝剂产生菌MBF4的絮凝活性分布在上清液中,而不是菌体细胞自身,通过分离提纯,最终得到微生物絮凝剂,经鉴定其不仅含有蛋白质类物质,而且还含有糖类物质。

【Abstract】 The study firstly screened a single strain capable of HNS wastewater-degrading which can produce MBF from HNS wastewater,the contaminated soil and poplar tree branches to be HNS wastewater, the complex bioflocculant with higher flocculation activity than MBF produced by a single strain were constructed. The optimize culture conditions and flocculation condition of the selected strain were ascertained through single factor experiment. The application of the complex bioflocculant in HNS wastewater treatment was examined in the paper. In the end,Flocculation active distribution was determined and chemical structural characteristics have been researched.15 strain were screened from HNS wastewater,the contaminated soil and poplar tree branches to be HNS wastewater, and the 7 strain with flocculant activity are screened. Those strain’s flocculation activity change in the process of domesticated. This phenomenon indicates that domesticated have an impact on the flocculant production. The complex bioflocculant with higher flocculation activity than MBF produced by a single strain were constructed, the flocculation could reach 77.60%.The studies show that carbon source for this strain to producing flocculant is soluble starch , optimum nitrogen source is peptone, optimal initial pH is 5.The studies show that the optimum flocculation condition is that the flocculants’ dosage is 3ml, pH value is 8, metal ion is Ca2+.The studies show that the optimum flocculation condition is that the flocculants’ dosage is 6ml, pH value is 8 in the HNS wastewater treatment. The ratio of removing chromaticity and COD of HNS wastewater-degrading is respectively up to 93.333percent and 73.08 percent.The complex bioflocculant are excreted out of the cell,and the flocculants are mostly distributed in the supernatant. The complex bioflocculant is not only sugar but also protein.
