【摘要】 目的：探讨三物白散大鼠含药血清对人胃癌SGC-7901细胞凋亡及抗凋亡基因survivin表达的影响。方法：选择人胃癌细胞细胞株SGC-7901,观察不同剂量组三物白散大鼠含药血清对SGC-7901细胞凋亡及抗凋亡基因survivin表达的影响。荧光染色检测细胞凋亡率,RT-PCR检测抗凋亡基因survivin的表达。结果：1.不同剂量组的三物白散含药血清干预SGC-7901细胞后,与生理盐水对照组相比,各组细胞的凋亡率升高,且细胞的凋亡率随三物白散含药血清浓度的增大、干预时间的延长,逐渐升高。2.不同剂量组的三物白散含药血清干预SGC-7901细胞后各组抗凋亡基因survivin的表达均有不同程度的下降,与生理盐水对照组比较,差异均具有统计学意义(P<0.05),且survivin表达随三物白散含药血清浓度的增大、干预时间的延长,逐渐下降。结论：1.三物白散在一定剂量、时间范围内,可诱导胃癌SGC-7901细胞凋亡,并呈一定的剂量、时间依赖性。2.不同剂量三物白散含药血清均能抑制SGC-7901细胞抗凋亡基因survivin表达,并具有剂量、时间依赖性。本研究结果表明三物白散有诱导胃癌SGC-7901细胞凋亡的作用,其机制可能与抑制抗凋亡基因survivin的表达有关。更多还原

【Abstract】 Objective:To investigat the effect of SanWuBaiSan-containing serum on the apoptosis and survivin expression of human gastric cancer cell lines SGC-7901.Methods:To choose the human gastric cancer cell lines SGC-7901, and SGC-7901 cells were treated by different concentrations of SanWuBaiSan-containing serum.The apoptosis was measured by fluorescence staining after the SGC-7901 cells were treated by SanWuBaiSan-containing serum,survivin expression of the SGC-7901 cells treated by SanWuBaiSan-containing serum were detected by RT-PCR.Results:1. After the SGC-7901 cells treated by different concentrations of SanWuBaiSan-containing serum.compares with the physiological saline control group, there was a increasing tendency on the apoptosis of SGC-7901 cells,and as the concentration increasing and time elongation,the apoptosis of SGC-7901 cells was increased gradually. 2.There was a decreasing tendency in different degrees on the survivin expression after the SGC-7901 cells treated by different concentrations of SanWuBaiSan-containing serum, Compares with the physiological saline control group, there was a statistics significance difference (P<0.05),and as the concentration increasing and time prolongation, the survivin expression of SGC-7901 cells was decreased gradually.Conclusions:1. SanWuBaiSan can induce apoptosis of SGC-7901 cells in a definite period of dose and time, and it depends on the definite dose and time.2.Different concentrations of SanWuBaiSan-containing serum can inhibit survivin expression of gastric cancer SGC-7901 cells, and expression with dose-dependence and time-dependence. The results of this study indicate that the SanWuBaiSan-containing serum could induce apoptosis of SGC-7901 cells and its mechanism may be related to inhibiting the expression of survivin.更多还原
