MMP-9基因多态性与复发性流产的相关性研究

【摘要】 目的:通过测定正常妊娠妇女和复发性流产史妇女的外周血的基质金属蛋白酶-9基因C1562T(MMP-9 C-1562T)多态性,拟从基因水平探讨MMP-9与复发性流产的相关性。方法:采用聚合酶链反应-限制性片段长度多态性( polymerase chain reaction-restrictive fragment length polymorphism,PCR-RFLP)分析法,对60例有复发性流产史的患者(病例组)和60例正常妊娠妇女(对照组)的MMP-9基因多态性进行分析,病例组及对照组年龄、孕周差异采用t检验;两组基因型频率及等位基因频率比较采用行列表资料的卡方检验分析,并计算表示相对风险度的优势比(odds ratio,OR)及其95%可信区间(confidence interval,CI)。结果:(1)病例组与对照组CT+TT基因频率分别为25%、10%,两组对比(χ~2=4.675,P﹤0.05)差异有统计学意义。(2)病例组与对照组T等位基因频率分别为12.5%、5%,两组对比(χ~2=5.483,P﹤0.05)差异有统计学意义。(3)与CC基因型和C等位基因相比,携带CT+TT基因型和T等位基因的人群能增加复发性流产的风险性,风险值分别为3.000(95%CI=1.075-8.370)、2.714(95%CI=1.015-7.255)。结论:(1)血清MMP-9基因多态性可能是导致复发性流产的危险因素。(2)T等位基因可能是复发性流产的遗传易感基因之一。

【Abstract】 Objective:By determining the normal pregnant women and recurrent spontaneous abortions history woman’s matrix metalloproteinases - 9 (MMP-9) gene polymorphism of peripheral blond to probe the relativity between MMP-9 gene polymorphism and recurrent spontaneous abortions.Methods:Using polymerase chain reaction - restriction fragment lengthpolymorphism (PCR - RFLP) analysis method to analyse sixty cases patients who have recurrent abortions history (case group) and 60 cases of normal health early pregnantwomen’s (control group) MMP-9 gene polymorphism , Using t test to checkout the difference of age, gestational weeks between the case group and the control group. Comparing two groups gene frequencies and allele frequencise uses the chi-square test(χ~2 ) analysis. Calculate odds ratio(OR ) to show relative risk degree,and its 95% confidence interal (CI) .Results:(1) The case group and control group CT + TT gene frequencies are 10% and 25% respectively,two groups’comparison (χ~2=4.675,P﹤0.05),the difference have statistically significant.(2) The case group and control group T allele frequencise are 12.5% and 5%, respectively, two groups’comparison(χ~2=5.483,P﹤0.05) the difference have statistically significant.(3)Compared with CC genotype and C alleles, carrying CT + TT genotype and T alleles crowd can increase the risk of recurrent miscarriage, value at risk (95% CI = 3.000 1.075-8.370), 2.714 (95% CI = 1.015-7.255) respectively.Conclusion:(1) MMP-9 gene polymorphism in serum may be the risk factors which cause recurrent spontaneous abortions .(2) T allele may be recurrent spontaneous abortions genetic susceptibility genes.
