山西道地药材黄芪HPLC指纹图谱研究及其药效调查

【摘要】 黄芪的主要化学成分包括皂苷类、黄酮类及多糖类等。2005版中国药典只规定了黄芪甲苷的含量,至今有关黄芪药材的研究很多,这些文献主要报道了黄芪甲苷、毛蕊异黄酮、芒炳花素的含量测定方法和黄芪药材指纹图谱的研究方法。色谱指纹图谱可以较为全面地检测其成分在药材中分布的全貌,使药材的内在质量情况可视。目前黄芪指纹图谱的研究主要有光谱法、色谱法、分子生物学方法等。目的建立山西省不同产地黄芪药材HPLC指纹图谱,为科学评价黄芪的内在质量提供参考。在山西省各个医院发放调查问卷,进一步验证山西四个产地的黄芪作为药材入药后对不同患者的治疗效果,同时了解山西四个产地的黄芪入药后在不同疾病中的疗效,以便于今后更好得为黄芪入药作指导。方法指纹图谱采用HPLC法,乙腈-水(30:70)四元梯度洗脱;流速0.8 mL/min;检测波长208nm;柱温:30℃。调查分析采用调查问卷形式,由山西中医学院06级中医系学生发出并且指导完成,调查数据采用卡方检验的统计学方法。结果山西省各个产地的黄芪药材中所含黄芪甲苷等成分均得到很好的分离,根据指纹图谱总结出了各个产地黄芪药材的质量区别。根据山西省各个产地的黄芪作为主要药材入药后不同患者的调查问卷,进行卡方检验,总结规律,可以显示出各个产地黄芪药材治疗各种病症的优劣,以便于更好得为今后黄芪入药作参考。结论1、由山西省四个产地的黄芪药材指纹图谱可以看出,山西浑源产黄芪所含黄芪甲苷组分的含量最高,所以在具体用药及实验中应优先选择山西浑源产黄芪。2、由山西省相同产地不同年限的黄芪药材指纹图谱可以看出,药材所含黄芪甲苷组分含量呈先上升后下降的趋势,所以采收黄芪药材时间最好在其生长期的第三、四年进行。3、根据山西省四个产地的黄芪作为主要药材入药后药效调查的问卷以及统计数据可以看出,在具体用药中应优先选择山西浑源产黄芪,其次为山西繁峙产黄芪。

【Abstract】 Astragalus main chemical compositions include saponins class, flavonoids and polysaccharides etc. The 2005 version of China Pharmacopoeia only stipulate the astragalus armour。Recently,there are many research on Astragalus.The literature mainly reported the Astragalus armour glucoside, calycosin separately, the awn polypropylene content determination of flower element method and Astragalus medicine fingerprint research methods. Chromatographic fingerprint can all-roundly detect its composition in medical herbs and make the inner quality be visible of visual. Currently astragalus fingerprint mainly have HPLC, molecular biology method, etc.ObjectiveEstablish different HPLC fingerprint for Radix Astragalus from different parts of Shanxi Province; Provide references for scientifically evaluating the inherent quality of Astragalus.We send questionnaires in several hospitals in Shanxi,in order to verify effect of four origin Astragalus medicine on patients, so that a better future for astragalus medicine for guidance.MethodsHPLC, acetonitrile-water (30:70) binary gradient elution; flow rate of 0.8 mL/min; detection wavelength 208nm; column temperature:30℃.Students of Shanxi TCM academy 06 class ZhongYi Department send and complete the questionnaire survey, survey data use chi-square test statistical methods.ResultsThe ingredients of Astragaloside from different parts of Shanxi Province were well separated. According to the fingerprint, the quality differences of Radix Astragali from each origin were revealed.According to different origin of Astragalus in Shanxi Province as the main medicinal materials medicine after different patients survey, to perform a chi-square test, and sum rules, can show each origin astragalus herbs treatment of various diseases of the advantages and disadvantages, so as to make better reference of Astragalus.Conclusion1、According HPLC fingerprint for Astragalus gathered from 4 different producing areas in ShanXi Provice, Astragalus,grown in Hun-yuan,contains the largest quantity of Astragaloside,and thus should have priority in clinical perscription and reseach esperiment.2、According HPLC fingerprint for Astragalus produced in the same areas but different producing time,the quantity of Astragaloside presents an increase first followed by decrease,and thus the best time for collection should be in the third year or the the fourth year after plantation.3、According to the questionaire and statistics, Astragalus produed in Hun-yuan ranks the first for clinical prescription and those produed in Fan-shi comesthe next.
