力竭运动后大鼠海马NF-κB、BDNF的表达变化

【摘要】 目的:以六周力竭游泳大鼠为模型,观察力竭运动后大鼠海马NF-κB、BDNF在不同时间点的表达变化,探讨力竭运动后中枢神经系统损伤及修复机制。材料与方法:健康雄性SD大鼠70只随机分为对照组(n=10)和力竭组(n=60)。力竭组进行6周力竭游泳训练后,选择即刻、4 h、8 h、12 h、16 h、24 h等不同时间点分别以冰上分离新鲜脑组织或多聚甲醛灌注固定后取脑,前者经Western-Blot半定量分析运动后NF-κB、BDNF的含量变化规律,后者用免疫组织化学SABC法染色,观察NF-κB、BDNF的表达变化规律。全部实验数据通过SPSS13.0统计软件分析处理,结果以(x|-)±s表示,以P<0.05为有显著性差异。结果:(1)Western-Blot结果显示,与对照组相比,力竭组海马NF-κB含量在4 h开始升高(P<0.05),8 h达峰值(P<0.05),12 h-16 h开始回落(P<0.05),24 h恢复到正常对照水平(P>0.05)。力竭运动后0 h组与对照组比较,BDNF的表达没有显著性差异(P>0.05),而4 h、8 h、12 h、16 h、24h组较对照组脑中BDNF表达明显升高(P<0.05)。(2)免疫组织化学结果表明,对照组海马NF-κB的表达以细胞胞浆为主,细胞核内偶见表达,即刻组NF-κB表达仍以胞浆表达为主,核内表达略多,两组间差异无显著性。4 h组细胞核内表达逐渐增多(P<0.05),并于8 h达高峰(P<0.05),随后12 h(P<0.05)、16 h(P<0.05)、24 h(P>0.05)各组核内表达逐渐减少。与对照相比,BDNF免疫阳性神经元则在24 h内持续表达且高于对照(P<0.05),12 h达峰值(P<0.05),12 h各组之间差异无显著性(P>0.05)。结论:①大鼠力竭游泳运动可以活化海马NF-κB信号系统,后者可能与运动性脑缺血再灌注时的神经细胞凋亡密切相关;②力竭运动诱导海马BDNF表达上调,从而产生相应的效应分子对神经元起保护作用。

【Abstract】 Objective:To observe the expression of NF-κB, BDNF in different times after 6 week exhausted swimming exercise . At the same time to investigate the mechanism of the injury and repair after exhausted swimming exercise in central nervous system .Methods: seventy healthy SD rats were randomly divided into control group(n=10) and exhausted swimming group(n=60).The fresh cerebral tissues were divided on icewater bath while the paraform-perfused brains were obtained at 0 h, 4 h,8 h,12 h, 16 h, 24 h after six-weeks exhausted exercise, the former to investigate the changes of NF-κB, BDNF in amygdaloid nucleus with western-blot method, the latter to observe the expression of NF-κB and BDNF by immunohistochemical SABC techniques.All of the experimental data were analyzed by SPSS13.0 statistical software, and expressed by (Mean±S.E.).Difference was considered statistically significant at P <0.05.Results:(1)Western-Blot results:compared with control group,the expression of NF-κB in hippocampus nucleus increased at 4 h(P <0.05),and peaked at 8 h(p<0.05), decreased at 12-16 h(p<0.05),and returned to normal levels at 24 h(P >0.05). The expression of BDNF at 4h、6h、12h、16h、24h were significant higher than the control group and 0h group(P<0.05). (2)Immunohistochemistry results: the expression of NF-κB was extremely lower and mainly resided in endochylema in control group and 0 h group,no difference was found between them. Increased and transfered into nucleus at 4 h after exhausted exercise(P< 0.05), and reached peak levels at 8 h(P< 0.05),and decreased at 12 h(P< 0.05),16 h(P< 0.05),24 h(P> 0.05) gradually.Compared with control group,the number of BDNF positive cells increased in all exercise groups dramatically(P< 0.05),exclude 12 h group reached peak levels (P< 0.05),and significant between other groups(P> 0.05).Conclusion: (1)The NF-κB signaling pathway can be activated in hippocampus nucleus in rats by exhausted swimming exercise. It is closely correlated with the inflammatory reaction and apoptosis during exhausted exercise.(2)Exhausted exercise could up-regulate the expression of BDNF,which might play an important role in protecting the neurons of rat hippocampus.
